| Abstract:Phosphorus is one of the most important nutrients for plant growth and development. Although there are a lot of phosphorus in nature, but most of them are in a variety of organic or inorganic insoluble phosphorus that can not to be absorpted directly. Therefore, high-yield crops at this stage still need to rely on a lot of phosphate fertilizer. But the application of P fertilizer in cropland is always susceptible to Ca2+, Al3+and Fe3+, etc. combine to insoluble phosphorus form, long-term application may result in soil compaction, and rainwater washed into rivers lead to eutrophication. Studies have shown that many plant endophytes on plant growth promoting effect, a part of them are Phosphate-solubilizing bacteria. Research on screening phosphate solubilization bacteria from the plants is not yet too much, especially the phosphate solubilization bacteria isolated from wheat was more rare. Promote Isolation and Screening of bacteria contribute to the expansion of dissolved phosphorus from microbial sources, phosphate solubilization ability of wheat endogenous endophytic bacteria repository for rich functionality and to develop new ways of improving soil phosphorus nutrition. And it has a positive and significant meaning for improving wheat yield, reduce the use of chemical fertilizers, protecting the ecological environment.The purpose of this study is obtaining the endogenous strains of wheat with high efficiency phosphate solubilization. First of all, collect the wheat plants from the normal cultivation of cropland, screening the strains with ability of dissolving the calcium phosphate from the root, stem and leaves of wheat plant. Study on its further dissolving characteristics, including an efficient phosphate solubilization endogenous strains named JG-22, through the observation of cell morphology characteristics, physiological and biochemical reactions and16S rDNA molecular biology identification showed that the strains of Brevibacillus agri strain.Research with the Brevibacillus agri strain JG-22fermentation of calcium phosphate, aluminum phosphate and aluminum phosphate, insoluble phosphorus source by changing the medium, pH, insoluble phosphorus content, incubation time, temperature of its melting ability of phosphorus.Through different media and culture conditions, Determination of dissolved phosphorus bacteria JG-22phosphate solubilization ability, JG-22solubility of phosphorus in calcium phosphate medium strongest, weaker ability to dissolve other insoluble phosphorus. JG-22in the initial pH, incubation temperature of34℃, dissolve the strongest. With the increase in insoluble phosphorus content in the medium, culture time lengthened, training to increase the content of available phosphorus in the liquid. Insoluble phosphorus content of5g/L, the incubation time of72hours after the content of available phosphorus in the culture medium with insoluble phosphorus content and the culture time increased significantly increase. |
