The golden cuttlefish (Sepia esculenta Hoyle) has many merits, such as rapid growing, wide feeding habits, high protein content and so on, it is a good species with high commercial value in north coastal of China. Up to nowadays, the studies on S. esculenta focuses on spawning migration, reproductive traits, biology characteristics and propagation characteristics, the researchs on growth rule and genetics is little. Therefore, for the purpose of protecting and making rational use of this resource, the fishery biology and basic research of genetics of the S. esculenta population of Qingdao, Shangdong Province, China, were investigated in this paper. In a sense, it is also has great significance for growth rule and genetics of the S. esculenta. In order to strengthen the chromosome preparation technology, this paper chooses the Synechogobius hasta as the object of the research, prepared the chromosome samples and analyzed its karyotype. The main research results are as follows:1. The relationship between beak shape parameters and mantle length, body weight of Sepia esculentaThe beak shape parameters and its relationship with mantle length and body weight were studied based on140beaks of Sepia esculenta which were collected from2009to2010. The results indicated that the upper beak of hood length and rostrum length, and the lower beak of hood length were best described by linear functions with mantle length but the beak dimensions showed positive exponential correlation with body weight. The equation between UHL and ML is ML=9.8770×UHL+4.7132, the equation between UHL, LCL and BW respectively are BW=41.716e01414UHL,BW=72.727e0.1376LCL. 2. Studies on the relationship between cuttlebone and growing of Sepia esculentaThe cuttlebone parameters and its relationship with mantle length and body weight were studied based on411cuttlebones of Sepia esculenta which were collected from2009to2010. The results indicated that the mantle length and cuttlebone length were best described by functions with body weight and striped number, the mantle length and cuttlebone width showed positive linear correlation with cuttlebone length; The equation between BW and ML are BW(larva)=5×10-3ML26713,BW(♀)=2.9×10-3ML2.2842,BW(♂)=9.6×10-3ML2.0877, the equation between cuttlebone width, mantle length, body weight and cuttlebone length are CL=2.8681CW-7.6930, ML(larva)=1.0541CL+0.4232, ML(♀)=0.9276CL+18.227, ML(♂)=1.0239CL+0.8035, BW=10-3CL25624, the equation between mantle length, body weight and Striped number are ML(larva)=137.3/(1+34.027e-0046SN), ML(♀)=208.253/(1+22.588e-0.033SN), ML(♂)=204.977/(1+35.246e-0.04OSN),BW=710.053/(1+434.682e-0.05OSN). Through the determination of cuttlebone length, cuttlebone width and striped number, combined with the functions of this research can accurate its mantle length and body weight.3. Chromosome Karyotypic Analysis of Sepia esculentaThe metaphase chromosome of Sepia esculenta was prepared from the oosperm, embryo cells by air dried. The karyotype has been analyzed. The results showed that there are92telocentric chromosomes in cell of S. esculentaand its karyotype is2n=44m+32sm+10st+6t, NF=168.4. Chromosome Karyotypic Analysis of Synechogobius hastaThe metaphase chromosome of Synechogobius hasta was prepared from head kidney cells by PHA and colchicine injection and hot air drying methods. The results showed that there are44chromosomes in Synechogobius hasta and its karyotype formula is2n=4m+40t, NF=48. Synechogobius hastabelongs to height classification in system classify and is the more primitive group.
