| Tomato was grown all over the world, and it was one kind of important vegetable of our country. But with the enlarged cultivating area and improper planting mode, the disease happened in tomato was more and more severity. A large of tomato have no yield because for Tomato yellow leaf curl virus disease break out.Breeding the resistant variety of tomato is best way of resistance to tomato yellow leaf curl virus disease.In this experiment, molecular marker technique was applied to investigate resistant genes of tomato yellow leaf curl virus in tomato,screen AFLP and SSR molecule marker linked to tomato yellow leaf curl virus disease resistant gene,establishing the AFLP and SSR molecule marker technique system of tomato,The results established the basis for resistance to tomato yellow leaf curl virus disease.The main results of this study is as following:1. In this study tomato resistant cultivar10032, susceptible cultivar10981and their hybridizing F1backcross BC1,BC2,F2progenies were used as the separating groups. It authenticated the resistance with bemisia tabaci inoculated,to establish resistance and susceptibility gene pools. The results indicated that all of F1groups were resistible, actually segregation ratio of F2groups was3.2:1and BC1groups was1:1, suggesting that the resistance gene was a single dominant gene.2. Using SSR and AFLP molecular markers,267F2population gained from10032×10981were used to construct the genetic linkage map. Three AFLP and one SSR markers linked to tomato yellow leaf curl virus disease resistant gene were identified in the study. And the Three AFLP markersE40/M49-Bã€E41/M49-Dã€E36/M48-E were3.4cM,11.6cM,18.3cM apart from Ty-3resistant gene differently. The one SSR markers SSR47was19.8cM away from Ty-3resistant gene.3. The AFLP molecule markers E40/M49-B was used to screen40tomato germplasm, and compare with Field identification,the inosculation rate is up to92.5%. |
