Eggplant (Solanum melongena L.) is one of the vegetables containanthocyanin. As a kind of widely spread natural pigment, anthocyanin hashigh capability of antioxidant activity, and has a wide range ofapplications with good market prospect. However, studies on eggplant aremainly focused on antioxidant activity evaluation and the key structuregenes screening. There is no report about anthocyanin biosynthesis sregulatory mechanism yet. The content and results of this study are below.1. MYB gene s cDNA and gDNA sequences in full length were clonedfrom eggplant cultivars YZ14and YZ3using RT-PCR and RACE.2. Sequence analysis shows that the full length of cDNA is1,035bplong, and the open reading frame(ORF)is837bp long, encoding278amino acids. The protein isoelectric point is at pH8.47. The SmMYBprotein contains two typical DNA-binding domains and has71% homologies with anthocyanin biosynthesis-related R2R3-MYBtranscription factor in capsicum. Furthermore, the protein is locatedmostly in the nucleus, in agreement with the regulatory function of MYBtranscription factors. The corresponding gDNA is3,834bp in length,consisting of three exons and two introns.3. For having accurate and credible data, we compared two samplingmethods of punching and slicing. The result by repeated experimentsshows that slicing method is more suitable to eggplant peel sampling.4. Fluorescence semi-quantitative PCR analysis indicates thatSmMYB is expressed in all organs of the plant, including roots, stems,leaves, petals, and peels. However, the expression level of SmMYB istissue-specific.5. The anthocyanin content and fluorescence semi-quantitative PCRanalysis after shading treatment indicates that the expression level ofSmMYB and SmDFR is highly correlated with the concentration ofanthocyanin in peel. Therefore, the SmMYB is speculated to be a MYBtranscription factor gene, which positively regulates the biosynthesis ofanthocyanin in eggplant.
