In this research, a whole genome scan was conducted on190progenies from afull-sib family using microsatellite markers and a half-sib mapping strategy wasemployed to identify QTL associated with activity of acid phosphatase in intestinal andalkaline phosphatase traits in both intestinal and liver. The main results and majorconclusion are as followings:1. Construction of genetic mapsIn the study,a genetic linkage map has been constructed for the common car usingJoinMap4.0software.315markers(28%) showed a significant distortion.The resultsshowed that the framework of genetic linkage maps consisted of992SSR markers in51linkage groups. The total linkage distance spanned by these markers was5183.9cM withan average of5.22cM for the whole framework map and the map coverage was88.8%.The biggest group was51linkage with9markers and the span was148.8cM;thesmallest group was1linkage with9markers and the span was42.2cM;the1linkagewith the highest high resolution was constructed with40markers spanning72.4cM andthe average was1.85.2.The location of QTL relation to activity of alkaline phosphatase traits in bothintestinal and liver and acid phosphatase in intestinal tissueFor the activity of alkaline phosphatase in intestinal,2QTL were identified at95%chromosome-wide level locating LG8(HLJ2634-HLJ3465) and LG13(CA2278-CA246)and explain the7.3%and7.0%variationFor the activity of alkaline phosphatase in intestinal,2QTL were identified at95%chromosome-wide level locating LG8(HLJ1244-2-HLJ1897) and LG41(CA642-HLJ495) and explain the20.1%and8.3%variationFor the activity of acid phosphatase in intestinal,5QTL were identified, of which1QTL was at the99%chromosome-wide level locating on the LG28and explain the39.9%variation;remaining4QTL were at95%of the chromosome-wide level locating on LG2(CA2049-CA2371), LG14(HLJ3275-CA174),(CA1276-CA2208)andLG25(HLJ2941-HLJ3946) and explain from8.1%to10%variation.3. Blasting the sequences of microsatellites locating both side of the QTL effectingactivity of ALP in liver to zebrafishTo search the candidate genes in the QTL regions which effect liver ALP producing,sequences of the mapped microsatellites locating both side of the QTL were used forsimilarity searches against genome sequences of zebrafish. The both side microsatellitesof QTL on LG8, HLJ1897and HLJ1244hit two loci (NW001877356.3and NW001877335.3) on the chromosome13of zebrafish. And the both side microsatellites ofQTL on LG41, HLJ3948and HLJ495also hit two loci (NW001877341.3and NW003040144.2) on the chromosome13. These two QTL corresponding area on thechromosome of zebrafish share a same region from31M to46M on the chromosome13of zebrafish.In corresponding share area, the protein phosphatase Mg2+-orMn2+-dependent (PPM) family was found. On the LG8corresponding area, norganicpyrophosphatase was found.This study constructed a medium density linkage map and identity QTL associatedwith activity of acid phosphatase in intestinal tissue and alkaline phosphatase traits inboth intestinal and liver.
