| The study was carried out to evaluate the effect of active immunization against adrenoceptor peptide vaccines which were produced through prokaryotic expression on growth performance and blood metabolites in rats, mouse and sheep. The results as follows:The Preparation of the peptide vaccines:The second extracellular loop of sheep α2A-AR and β2-AR E.coli expression vectors were constructed through DNA recombination technique, which could express in the E.coli BL21(DE3) by inducing of IPTG, which provide basics for producing cheap adrenoceptor peptide vaccines. The result showed that relative molecular masses of recombination α2A-AR and β2-AR peptides were consistent with the expected25.3and27.39KD. The proportion of α2A-AR peptide was0%,25.0%,46.7%,61.7%and60.5%of total bacterial protein after being induced by0,30,60,90and120min. That of β2-AR peptide was0%,26.4%,40.3%,59.2%and60.0%. After being purified by affinity chromatography, the proportions of α2A-AR and β2-AR accounted for total protein were97.4%and97.7%respectively.The effect of peptide vaccines on Growth Performance and of Blood Metabolites in rats:Eighty female SD rats were randomly allocated to five groups, i.e. the control group, CL group, α2A-AR group, β2-AR group and SP group. The a2A-AR group,β2-AR group and SP group were active immunization against the corresponding antigen at the beginning of the second week (70μg/rat) and boosted at the fourth week (35μg/rat). The CL group was fed with the diet containing clenbuterol (3mg/kg) at the beginning of the third week. Saline were injected into the control and CL group at the same time of the first and booster injection,respectively. The rats were given free access to feed and water and intake was measured every week. The results showed that high level antibody was produced by rats after peptide vaccine immunization, but that of SP group was low. It was significant influence on body weight and slaughter performance of rats after being active immunization of peptide vaccines. To compared with control group, the ratio of abdominal fat and TG were reduced by76.74%(P<0.05) and28.13%(P<0.05) by CL administration. Compared to the control, however, the concentrations of TG and TC of the β2-AR group were improved by34.15%(P<0.05) and22.94%(P<0.05) than control group. The concentrations of GLU and TC were raised by35.45%(P<0.05) and20.59%(P<0.05) in the SP group.Effects of active immunization against adrenergic receptor peptide vaccine on growth and blood metabolites of female mice:One hundred female KUNG-MING mice were randomly allocated to five groups, i.e. the control group, CL group, α2A-AR group,β2-AR group and SP group. The α2A-AR group, β2-AR group and SP group were active immunization against the corresponding antigen at the beginning of the second week (30μg/mice)and boosted at the fourth week (15μg/mice). The CL group was fed with the diet containing clenbuterol (3mg/kg) at the beginning of the third week. Saline were injected into the control and CL group at the same time of the first and booster injection, respectively. All the mouse were given free access to feed and water and intake was measured every week. The result showed that the slaughter rate of CL group was higher than others groups, and the ratio of mesentery was significant lower than that of others group, and the ratio of leg muscle was higher than that of others groups. Compared with control, the concentrations of TG and LDL-C of CL group was reduced by17.69%(P>0.05)12.35%(P>0.05). Intake, body weight and slaughter performance of a2A-AR group,β2-AR group and SP group. The concentrations of LDL-C of the α2A-AR group and the SP group were lowered by19.74%(P<0.05) and12.35%(P>0.05) than the control group.Effects of active immunization against adrenergic receptor peptide vaccine on growth and blood metabolites of sheep:Twelve male sheep with body weight31kg were randomly allocated to two groups, the control group and β2-AR group. The sheep was active immunization at the first day and was boosted at the fifth day. The sheep were given free access to feed and water and fasting body weight was measured every fifty days. Blood samples were drawed from jugular vein four times with interval of15min before feeding and after feeding respectively. Ninety days later, carcasses of the all sheep were measured and the serum was used to test antibody level and biochemical metabolites. The result showed that active immunization of β2-AR peptide had no influence on intake, body weight and biochemical index after feeding. The concentration of GLU of experiment group was higher21.74%(P<0.05) than that of the control group before feeding at the fifth day, and the concentration of LDL-C was reduced by15.19%(P<0.05) before feeding at the sixtieth day; to compared with control group, the slaughter rate of the experiment group was improved significantly4.60%(P<0.05).To sum up, rat, mice and sheep were active immunization against peptide vaccines could produce antibody and improve sheep’s slaughter rate, but it is no influence on the growth and feed intake and fat deposition of rat, mice and sheep. |
