Jujube is a plant of Ziziphus Mill.of Rhamnaceae. Production holds in the first place in the three big dried fruit. Jujube flowering is inconsistent, and the flowers are small and pollination is very difficult. Under natural conditions jujube has lower fruit-bearing rate and higher embryo abortion rate, meanwhile has serious diseases and insect pests. The traditional methods of breeding cycle is long and low efficiency, and it is hard to to get a good resistance new varieties. The experiment was based on the material of mature embryos of sour jujube,and stablished dates from the body embryo regeneration system. It’s a foundation of embryo research for jujubes genetic transformation. In addition, the study was based on the material of mature embryos of sour jujube and the leaves of Z. jujuba ’Huizao’ to research the jujube agrobacterium mediated insect-resistant genes genetic transformation. The main results were as follows:1Establishment of mature embryos regeneration system in jujubeThe results showed that the higher germination rate of mature embryos was kept a year at normal temperature on MS without any hormones. And the best proliferous medium was1/2MS+6-BAl.Omg·L-1+NAA0.3mg·L-1, meanwhile a better braking effect on browning was added PVP l.5g·L-1into the medium.2Study on agrobacterium-mediated genetic transformation of jujubeThe study was based on the material of mature embryos of Z. jujuba ’Fengmiguan’and the leaves of Z. jujuba ’Huizao’. It’s hard to take off the bacterium after using EHA105strains agrobacterium-mediated to infect explants. Only when the bacterium fluid concentration was in the OD600=0.2, it can be effectively antibacterial and get in vitro shoot. Jujubes were very sensitive to herbicides basta, and the browning mortality was as high as96.5%even the concentration was in2mg/L. It was considered that herbicides basta should not used as screening agent in genetic transformation for jujubes.
