| Duck Hemorrhagic Ovaritis(DHO) is a Infectious disease caused by a new type of flavivirus, making infected duck declined and had a rate of sudden drop in egg production as the main features. In this study, through infected SPF ducklings and SPF chickens with DHOV, we established the Duck Hemorrhagic Ovarian artificial infection model, study the clinical symptoms, the pathological changes, the distribution and positioning of the viral antigens, route of transmission and etc, in order to determine this disease typical symptoms, lesion characteristics and pathogenesis clearly, to provide the experimental basis for the prevention and control of disease in poulotry production. In this study, twelve SPF ducklings34days of age and eleven SPF chickens120days of age were inoculated by the oral route of F4generation DHOV-HB strain respectively; Animals in the control group vaccinated duck embryo allantoic of the same dilution degree and the same dose. After inoculation animals were observed their clinical symptoms,and collected nasopHaryngeal swabs to inoculate chicken embryos for virus isolation, on different days after infection dissected SPF ducklings and SPF chickens pathologically, at the same time take various tissues fix in formalin、the slices were done for observing histopathologically and virus immunohistochemical staining. The results showed that, the typical symptoms of SPF ducklings’infection:poor appetite, pulled the green loose stools, the particularly bad spirit, liked lying and got together; grossly the pathological changes were that spleen and kidney significantly enlarged, there were bleeding spots on the liver surface; microscopicly the pathological changes were that inflammatory cells infiltrated in the spleen, dieldrin cavity showed in the liver, sinusoidal endotheial cells and Kupffer’s cells were mast、activated and proliferated, interstitial nephritis, non-suppurative encephalitis, islet dissolved, etc. Viral titers of virus in liver was the lowest. Viral antigen located in the cytoplasm of a variety of cells, the spleen located in the reticular cells and lymphocytes, kidney located in the tubular epithelial cells, liver located in the liver cells and sinus wall cells, heart located in the myocardial cells, pancreas located in the islet cells, thymu located in the reticular cells. SPF chickens inoculated were no obvious clinical symptoms and pathological changes, the virus re-isolated were negative. This experiment established DHO artificial infection model of SPF ducklings. |
