Recent studies have found that inhibin is affected sheep lambing number of important genes,-this experiment with inhibin as candidate genes, from gene polymorphism to quantitative expression combined with Bamei mutton sheep lambing number do related research.This paper adopts lambing number of sheep blood sampling record Bamei mutton sheep, detected by PCR-SSCP INHβA and INHa gene in sheep and Mongolia sheep and the Bamei mutton sheep on gene polymorphism, genotype and lambing number of sheep to make correlation analysis.Collecting3consecutive births with single or twin lamb lamb production records and the Bamei mutton sheep uterus and ovaries, with RT-PCR method on INHβA and INHa subunit gene relative quantitative detection, further analyzed by single and twin lamb lamb by expression differences and lambing number of relations.The results found that primer1,2,5and7amplified fragment polymorphism is, the remaining three pairs of primers amplified fragment was not polymorphism. Primer1, Bamei mutton sheep have three genotypes (AA, BB and AB), primer2, and Bamei mutton sheep two variety of genotype CC and CD type, CD type CC than typeO.17(P<0.01). Primer4, and Bamei mutton sheep in three genotypes (EE, FF and EF), the average number of lambs and sheep type FF than type EE multiple of0.44(P<0.01). Primer6, and Bamei mutton sheep have three genotypes (GG, HH and GH), type GG than type HH average lambing numberO.21(P<0.01), the results show that, inhibin alpha subunit (INHa, INHA) and beta subunit A (inhibinβA, INHBA) genes are Bamei mutton sheep Lambing number of important genes.Through the RT-PCR method for the detection of INHβA and INHasubunit gene in Bamei mutton sheep oestrus after1hours,12hours and24hours time point of uterine and ovarian tissue in the relative expression levels, and expression differences. Bamei mutton sheep oestrus after1hours,12hours,24hours, and INHβA and INHa subunit gene in ovarian expression of quantity increased first and then decreased. In the period of ovulation inhibin expression of inhibin in larger, uterus and ovary of expression changes were not synchronous, expression in the uterus weight change with the ovarian changes, but there is a time delay. Experimental samples by twin lamb sheep after estrus than multiparous Bamei mutton single lamb sheep ovulation and the Bamei mutton sheep in advance, advance into the next estrous cycle, and ovulation number than multiparous single lambs. |
