This experiment investigated the protective effect of Gln on Aeromonas hydrophila infection of Pelodiscus sinensis. two hundred and seventy healthy Pelodiscus sinensis with an average weight of (220.75±12.65) g were selected, and randomly divided into5groups. GroupA,B,C,D injected with Aeromonas hydrophila (1.3×109cfu/mL,0.5mL) in order to cause the disease, the Group A and E was injected with normal saline and then the other three groups were injected with100,200,400mg/kg of glutamine injection0.5mL after the pathological symptoms appeared,. Injected once every two days, a total of three injections. Sampling in the7d the the first injection the Gin, to observe in each group the survival rate、Antioxidant capacity、Non-specific immune function、 Histological changes of the liver, spleen and intestine、Intestinal microflora of the Pelodiscus sinensis. The results are as follows:(1). The survival rate of group A is very significantly reduced of group E (P<0.01), the survival rate of group D is significantly increased of group A (P<0.05)。(2). Compared with group A, CAT, GSH-PX levels of Group B were very significantly increased (P<0.01), SOD levels were significantly increased (P<0.05), the MDA level significantly reduced (P<0.05); CAT, GSH-PX,SOD levels of Group C were very significantly increased (P<0.01), the MDA level significantly reduced (P<0.05); CAT, GSH-PX,SOD,T-AOC levels of Group D were very significantly increased (P<0.01), the MDA level significantly reduced (P<0.05); Group D compared with Group E, the T-AOC, CAT and GSH-PX were not significant in the differences (P>0.05); that Gln plays an important role of antioxidant capacity with Pelodiscus sinensis infected with Aeromonas hydrophila.(3). Compared with Group A, group B red blood cell phagocytic rate significantly increased (P<0.05), leukocyte phagocytic rate was very significantly increased(P<0.01). lysozyme difference was not significant (P>0.05); Group C, lysozyme, leukocyte phagocytic rate and red blood cell phagocytic rate increased significantly (P<0.01); group D, leukocyte phagocytic rate and red blood cell phagocytic rate increased significantly (P<0.01), the survival rate significantly increased (P<0.05); and the lysozyme of the group D compared with Group E, the difference was not significant (P>0.05); that Gln can improve Pelodiscus sinensis infected with Aeromonas hydrophila a variety of immune factors level, thereby enhancing the body of non-specific immune function.(4). Pelodiscus sinensis infected with Aeromonas hydrophila liver manifested as severe congestion, a large number of red blood cell infiltration in the liver cell, the gap of liver cell increases, hepatocyte nuclear was degeneration, with the increase of injection volume, liver bleeding point decrease, liver bleeding on a decreasing trend, reduce empty cells, liver cells, body size, arrangement tends to be normal; intestinal lesions showed intestinal epithelial shedding, intestinal mucosal atrophy, intestinal micro bleeding, after injected with Gln the width of the intestinal mucosa was increased, intestinal villus morphology tended to be normal;After infected with Aeromonas hydrophila Pelodiscus sinensis spleen capsule vascular swelling, red blood cells clearly inside,white pulp is compressed, lymphocytes and blood cells of the red pulp and white pulp necrosis, injection of Gln, red blood cells in the spleen reduced, red and white pith became clear boundaries, cell structure gradually became normal. So,That Gln can contribute to Aeromonas hydrophila infection of the intestine, spleen and liver of the Pelodiscus sinensis recovery.(5). Group A Aeromonas hydrophila,E. coli was significantly higher than Group E (P<0.05), Groups C and D Aeromonas hydrophila and E. coli were significantly lower than group A (P<0.05), Gln could improve the number of Bacillus and Lactic acid bacteria in intestine significantly (P<0.05), Bacillus and Lactic acid bacteria of Group A was significantly lower than the Group E (P<0.05), Bacillus and Lactic acid bacteria of Group B,C,D were significantly higher than the Group A(P<0.05). So Gln could improve the number of beneficial bacteria and reduce the number of harmful bacteria of Pelodiscus sinensis infected with Aeromonas hydrophila.To sum up:Gln significantly improved antioxidant activity and the non-specific immunity of Pelodiscus sinensi infected with Aeromonas hydrophila. Good for the promotion of intestinal bacteria colonization and restoration of liver, spleen and intestine,increase the survival rate.So,Gln has a protective effect of Pelodiscus sinensi infected with Aeromonas hydrophila.
