| Chinese jujube (Ziziphus jujube Mill.) originated from China. Gremplasm resourcesof Chinese jujube are very abundant. The classification evolved from morphologicaldiversity to genetic diversity. The studies on Ziziphus jujube classification is very limited,the synonym and homonym are very seriously presented. In addition, the geneticrelationships among cultivars were not very clear. In this study genetic diversity in85Chinese jujube accessions from different regions was assessed Using ISSR makers. Thencore collection of Chinese jujube was developed. The main results were as follows:1ã€Finally establish an optimal reaction system of PCR for Chinese jujube which is: ina25ul PCR reaction system contains1.0ul Taq DNA polymerase,2.5mmol/L MgCl2,0.20mmol/L dNTPs,0.30umol/L primers and20-80ng template DNA. And the bestresponse system is validated. Mg2+and dNTPs are the two main affective to ISSR-PCRreaction.2ã€26informative primers selected from32primers were used in ISSR analysis, basedon level of polymorphisms and the number of strong bands. A total of255bands werescored,of them240(94.12%) were Polymorphic. The genetic similarity (GS) among85accccessions ranged from0.4863to0.9490. The genetic similarity betweenFeidongyuanzao and Feidonglingzao is the highest (0.9490). On the other hand,Thegenetic similarity between Xuanchengyuanzao and Tongqianshu, The genetic similaritybetween Shandonglizao and Tongqianshu are the lowest (0.4863).3ã€Cluster analysis using UPGMA (Un-weighted Pair Group Mathematics Average)method based on the data of ISSR. The clustering results have a close relation to thegeographic location of the date seedling resources.4ã€The results of amplification analysis of cultivars of Anhui province show that thetotal of223bands were scored,of them190were polymorphic, the percent polymorphicloci is85.20%. The genetic similarity (GS) among34accccessions ranged from0.6392to0.9490. The genetic similarity between Feidonglingzao and Feidongyuanzao is the highest(0.9490), the genetic similarity between Fanchangxiaodongguazao andWannandongguazao is secondly(0.9373); On the other hand, The genetic similaritybetween Fanchangxiaodongguazao and Xuanchengyuanhao is the lowest (0.6392), andthen are the genetic similarity between Wannandongguazao and Xuanchengyuanzao, is0.6549.5ã€It is the first time to research the genetic diversity of groups in Anhui province and the surrounding areas. Compared the parameters such as the number of polymorphic loci,percentage of polymorphic loci, and observed the number of alleles, effective number ofalleles, Nei’s gene diversity and Shannon’s information index of2groups (AnHui and TheNorth).
