Trichinosis is a zoonotic disease with worldwide distribution and which causesmainly by eating raw meat containing infective muscle larvae of Trichinella..Trichinella spiralis has a wide range of spices, at present,Trichinella compriseseight species and four genus in the world, involving encapsulating andnon-encapsulating Trichinella, among these Trichinella pseudospiralis(T4) is anon-encapsulating Trichinella.Recent years, the infection of Trichinella pseudospiralis is growing increasinglyserious and even occurred outbreak and epidemic in human, for example, the firstpandemic of T.pseudospiralis in human history occurred in the southern region ofThailand from1994to1995, and in France also had a human T.pseudospiralisoutbreak due to the consumption of wild boar in2000, which caused a huge threat tohumans, thus the study of T.pseudospiralis is becoming particularly important.Our laboratory successfully constructed the muscle larvae cDNA library ofT.pseudospiralis by Wu et al and had an immunological screening, which obtained ahigh specificity and strong antigenic antigen gene WR29, encoding proteasomeactivator subunit PA28β, and it is a specific protein secreted by T.pseudospiralismuscle larvae by verification. Some studies have shown that PA28β played animportant role in immune response process, so we predict that WR29also have someimpact on the host immune, therefore we made some preliminary study on it.There are three parts in this experiment, first, the WR29recombinant plasmidwhich preserved in our laboratory were identified and induced expression, in order toprepare recombinant protein. After purification and identification, the recombinantprotein were injected to the rabbits to get antiserum, the results showed thatpurified protein is almost non-miscellaneous band, and the titer of immune serumreached1:12800; Second, we collected T.pseudospiralis (isolated from Romanian pig,international standard insect ID ISS013) muscle larvae excretory-secretoryproducts(ES), using the antiserum from the first part of the experiment extracted thenatural WR29protein from ES by Immunoaffinity Chromatography method, while made use of Western Blot for identification, the results reveled that the extraction ofprotein had a higher concentration, Western Blot method showed that the protein hadreaction with swine infected with T.pseudospiralis60days; Finally, the extractedprotein were used for animals experiments, to detect the secretion of cytokines fromserum and spleen, the results showed that the content of the anti-inflammatorycytokine IL-10, IL-4and TGF-β increased, while pro-inflammatory cytokine IFN-γdecreased. Currently, the study for the interaction between T.pseudospiralis and hostis so limited that we are unclear the fuction of WR29, thus need for further study. Inthis study, it promoted the establishment of specific and sensitive T.pseudospiralisdiagnosis antigen and diagnosis method.
