Potentilla Chinesis Ser. belongs to the Rosaceae Potentilla. Because of itsreliable effects, it is prescribed in every edition of 《Chinese Pharmacopoeia》.There are many species of Potentilla, only in Jilin province there are over tenspecies. Due to their close relationship, some of the plants have similarities inmorphology; and no matter they live in the same environment or not, theirchemical components are the same, therefore, using the ways of morphology,microscope, TLC and HPLC to identify them is very difficult. In addition, thetraditional ways have many influential factors which lead to changes in theirmorphology, histological structures and chemical elements and influence theaccuracy of the identification method itself. Molecular identification is stableand not easily influenced by the temperature, mechanical force and growingenvironment. Because of its convenience, quickness, stability and goodreproducibility which are different from traditional methods, it has become anew identification method. The fundamental of these methods is the DNAsequence in specific region of the plants that are to be identified. So sequenceanalysis becomes the basis of the molecule identification.Based on this perspective, in this study we abstracted the genome DNA of5common Potentilla collected in Jilin province; amplified the region of ITS andTrnL-F using the universal primer of the angiosperm; and then purified theamplified products and sequenced them directly. The results shows that theITS or TrnL-F sequences of the same species of Potentilla are same, butsignificant different between either two species of Potentilla, so ITS and Trnl-Fsequence analysis and methods derived from them can be used inauthentication of Potentilla.
