ObjectiveTo build animal models of the mice that infected with Eperythrozoon from pig,detect the variation condition of the mice’s peripheral blood infectedEperythrozoon BALB/c and correlation factor CD35, CD58and CD59which areimmune to red blood cell, also studying CD2molecules of the T cell membrane.In order that, explore the red blood cell have infected the Eperythrozoon fromanimal model how to play the immune function. To provide the theory basis forascertaining have infected Eperythrozoon how to cause the inflammatoryresponse and the immune Response Mechanism.MethodAseptically collected40portion pig blood for the animal husbandry of Lao Ningmedical school. Identified positive Eperythrozoon blood by microscopicexamination and PCR. Randomly divided mice BALB/c to the infectedEperythrozoon group, the negative group, the control group. The three groupsinoculated respectively0.5ml positive blood samples of pig’s Eperythrozoon,0.5ml negative blood samples of pig’s Eperythrozoon,0.5ml normal saline.After the mice are inoculated, respectively extracted blood in mice at1d,3d,5d,7d,9d. Defect variation of CD35on the surface of the mice red cell and CD35,CD58, CD59in blood plasma by ELISA. Defect CD58, CD59on the surface ofred cell and CD2on the T cell by flow cytometry. Result1.ELISA show: After BALB/c mice infected eperythrozoon,1std、3rdd、5thd、7thd、9thd after,measured CD35on the surface of red cell present decreasetrend gradually, lower the control group (P <0.05). However, CD35, CD58,CD59in blood plasma present increase trend, they reached the peak at the thirdday, they were higher than other control group, the difference was statisticallysignificant (P <0.01).2.Flow cytometry show: After BALB/c mice infected eperythrozoon, Erythrocytemembranes of CD58and CD59positive percentage showed a decreasing trend,significantly lower than the other groups (P<0.5), CD2-positive rate increased,and higher than the other groups(P<0.5).3.Positive group of CD58、D59and CD2-positive percentage of statisticalanalysis, found that CD58and CD2, CD59and CD2show negative correlation,r=-0.726(P<0.5), CD58and CD59shows positive correlation r=0.723(P<0.5).Conclusion1.Using Eperythrozoon to infect BALB/c mice, the form and structure oferythrocyte surface changes. At the same time, it affect expression of CD35、CD58and CD59of erythrocyte which related erythrocyte immune.Thus,recognition and adhesion-function of the erythrocyte is blocked, affecting theimmune function of T cell and B cell.2.Using Flow cytometry to check the expression of erythrocyte mem-branes’CD58、 CD59and T lymphocyte’CD2. It confirm the erythrocytemembranes of CD58and CD59reduce when mice are infected byEperythrozoon, and are not able to combine, consequently the immune oferythrocyte is affected; another, the shedding of CD58has ability to bind withthe CD2after erythrocyte membranes have damaged, and regulate the immune faction of T cell.
