This study use wheat mutants10-HN20mediated by the ion beam of soybean DNA sixth generation stability (easy to aging,file as C),10-HN30(anti-aging, file as D) and new wheat9(controlled experiment,file as CK) as material to research nucleus of different cell types (long cell, shortcells, guard cells, accessory cell) and guard cell chloroplast dynamics on grain filling in wheat flag leaf during senescence by using fluorescence microscopy, and analyse POD zymogram on grain filling in wheat flag leaf through renaturation electrophoresis technology. aiming to explain the senescence of wheat leaves during senescence of different cell types at the cellular level and the aging dynamic changes of antioxidant POD associated with aging, and provide a theoretical basis for further study of wheat late senescence mechanism and cultivation of anti-aging new product.This results show that:1. Aging speed of Wheat variant strains of10-HN20,10-HN30and late reproductiveepidermal cell of collation is:10-HN20> CK>10-HN30, and consist with the aging of external morphology.2. morphology of aging epidermis cell is diversification, appears oval, crescent shaped, bar, cometary, and dispersed and so on, disintegration rate of different types of cell show asynchrony, the speed of disintegration is: long cells> short cells> guard cells> subsidiary cells.3.disintegration of chloroplast and nuclear inguard cell is asynchronous when leaf is senescence, and chloroplasts disintegrate early in the cell nucleus.4.in leaf senescence period, the number of disintegration of chloroplasts of guard cell in three wheat lines is:10-HN20> CK>10-HN30;5.10-HN20,10-HN30variant is significantly different POD isozyme bands of controlled experiment in filling stage, the number of enzyme belt and total enzyme activity is relate to material aging degree, manifesting that there are many anti-aging lines10-HN30isozymes band, and total enzyme active is better; enzymes band of lines10-HN20which is easy to aging have the least number,and total enzyme activity is weak.
