Collecting healthy capsicum plants in Qing Shui and Meng Da country ofXunhua county,19strains of bacteria were obtained by using dilution-plate method,among15strains were endophytic bacteria after inoculating them into plants.Thestrain Yg-3was screened for their stable antagonist and growth promoting effects.By studying the biological characteristics of strain Yg-3,it was shown that itwas Gram positive(G+) Bacillus,the colonial of morphology is semi-yellow,non-regulated edge,was jagged or wavy,rough surface,opaque,with the culturetime prolonged,becoming a colony may have folded into brown-like uplift,hascircumjacent flagella. Combination of colonial and physiological and biochemicaltest results,Yg-3may be initially identified as Bacillus Subtilis.Strain Yg-3is inserted into capsicum by three ways: irrigating roots,dippingseed and injecting,endophytic colonization study have shown that the colonialamount of irrigating roots is the largest. Only is the colonial amount of leaves at theoptimum,there is no marked strain in the root and stem.The results of soaking in different time that colonial amount is increasing whenprolonging soaking time,the colonial amount of24h is the largest. Soaking indifferent concentrations of bacillus colonization measurement results shows that thecolonial amount is at high level by diluted ten times and the colonial amount isreducing with concentration of bacillus colonization. This shows that the differentsoaking time and concentrated soaking for different strain to enter the plant musthave influenced. The strain Yg-3also can live and move in the different capsicum,so the strain have no selectivity for capsicum breeds.By fermentation craft shows that the best culture medium recipe of Yg-3is thefourth,Soy flour30g,Gluate20g,Starch5g,Yeast extract2g,NaCL4g,K2HPO40.5g,MgSO4·7H2O0.5g,CaCO32g,H2O1000mL. The ultimate biosynthesis willbe achieved when the pH6.5,the temperature at30℃,the concentration is1%, rotated speed is150r/min,for strain Yg-3.The different treatments (1×109,1×108,1×107,1×106CFU·ML-1)of strain Yg-3totreat the seeds and young plants of capsicum,the test showed that the fermentedliquor have inhibiting effect for it. seed germination,dry weight,the length of rootand stem,emergence rate,fresh weight of pepper,on the contrary,the dilutionhave promoting effect in some degree.Fermented1×108CFU·ML-1for Yg-3 is themost obvious in it. The best effect has appeared in the first way,highly significantcontrol with difference,the second is dipping seed,significant control withdifference, the last is injecting. Strain Yg-3was mixed sheep manure fermentationseed soaking,seed dressing mixed with grass charcoal,bacterial seed soaking to thelength of root and stem,emergence rate,fresh weight of capsicum have inhibitingeffect.
