Reddish-brown raccoon dog is a kind of mutation raccoon dog. Hair trip of reddish-brown raccoon dog is reddish-brown and to menus is white. Reddish-brown raccoon dog’s eyes and lips are pink. The new type of mutation hair color is controlled by explicit heterozygous gene. The research about checking SNP of MC1R gene is carried out in order to process the inheritance base of mutation hair color.MC1R which is composited of317entries amino acid and7sequence across membrane is dynorphine.MC1R which is a kind of G-protein homo-coupling receptor is called melanin cell hormone receptor.MC1R combining endogenous ligand or Agouti protein acts important part in the process which melanin cell makes melanin, which is of great importance about control hair color.In the experiment for research object, that had8wild Wusuli raccoon dog,7reddish-brown Wusuli raccoon dog,6Jilin white raccoon dog and6silver-black fox from Qingdao Lacy breeding base. By the SNP testing, we can analysis the correlation of MC1R gene expression level in the Wusuli raccoon dog in different colors.As known as NCBI sequence (GenBank:HM852533.1), we designed primer, sing for getting MC1R gene which play an important role in mammals melanin cell synthesis melanin process. With the extraction of raccoon dog hair follicle DNA as a template, PCR amplification was successful to get the reddish-brown raccoon dog MC1R gene sequence for1329bp, and the known sequence homology91.47%,18mutations; with the wild type Wusuli raccoon dog homology reached99%,8mutations. Testing to eight variation, respectively274bp,439bp,452bp,474bp,569bp,793bp,820bp,829bp,53site of the439bp coding pralines third antic don mutations, from A to G mutations, belong to no righteous mutations. The rest are amino acid sequence to change, namely. Non-cSNPs were included Gln274Arg, Va1452AIa, Arg474Leu, Arg569Cys, Arg793Cys, Arg820His, and Ala829Glu. Through the enzyme cut in MC1R genes, two genes were got enzyme type AB and BB type about300bp, reddish brown raccoon dog for AB, wild type raccoon dog for BB type. Through the real-time PCR analysis results showed that the quantitative:expression of reddish brown Wusuli raccoon dog with wild type Wusuli raccoon dog, Jilin Bai raccoon dog, silver-black fox’s hair follicles is consistent.
