OBJECTIVE:Bovine mammary epithelial cells and bovine mammary fibroblasts cells were cultured and purified in the study. Biological characteristics of these cells were identified by observing cells in chateliertype microscope and drawing cellular growth curves. To expound the effects of TGF-Pi and bFGF on proliferation and apoptosis of bovine mammary glandular cells.METHOD:Bovine mammary cells were cultured by tissue block method and single cell isolation method. On the basis of the degree of bovine mammary epithelial cells and bovine mammary fibroblasts cells anchored to a surface were different. The bovine mammary epithelial cells and bovine mammary fibroblasts cells were purified by digestion with different concentrations trypsin for5-6passage times. The shape of cell was observed by chateliertype microscope. The source of the cells were identied by the method of immunohistochemistry. The effects of TGF-β1and bFGF on proliferation and apoptosis of bovine mammary glandular cells were detected in the study.RESULT:(1) The result show that the cells have typical morphological features have good ability of proliferation.(2) Experimental groups with different concentration of TGF-β1were significant different compared with the control group. The effect of inhibition cell proliferation was enhancement with the increase of the concentration of TGF-β1in bovine mammary epithelial cells; Experimental groups with different concentration of TGF-β1were significant difference compared with the control group, the effect of promote cell proliferation was enhancement with the increase of the concentration of TGF-β1in bovine mammary fibroblasts cells by the method of MTT. Experimental groups with different concentration of bFGF were not difference compared with the control groups in bovine mammary epithelial cells; Experimental group with different concentration of bFGF were significant difference compared with the control groups, the effect of promote cell proliferation was enhancement with the increase of the concentration of bFGF in bovine mammary fibroblasts cells by the method of MTT.(3) Experimental groups with different concentration of TGF-β1were significant difference compared with the control groups, the effect of promote apoptosis was enhancement with the increase of the concentration of TGF-β1in bovine mammary epithelial cells; Experimental groups with different concentration of TGF-β1were significant difference compared with the control groups, the effect of inhibition apoptosis was enhancement with the increase of the concentration of TGF-β1in bovine mammary fibroblasts cells. experimental group with different concentration of bFGF was not difference compared with the control group in bovine mammary epithelial cells; Experimental group with different concentration of bFGF were significant difference compared with the control group, the effect of inhibition apoptosis was enhancement with the increase of the concentration of bFGF in bovine mammary fibroblasts cells.CONCLUSION:The result show that proliferation of bovine mammary epithelial cells were inhibition and the apoptosis of the cells were promote by TGF-β1; Proliferation of bovine fibroblasts epithelial cells were promote and the apoptosis of the cells were Inhibition by TGF-β1; The result show bovine mammary epithelial cells were not Influence by bFGF; The result show that proliferation of bovine fibroblasts epithelial cells were promote and the apoptosis of the cells were inhibition by bFGF.
