This stud adopts16x6NC Ⅱ design, used the96hybrid corn and22parent material, to using cDNA-SRAP molecular markers in the reverse transcription level for the first time, its parent of hybrid corn kernels expression pattern of genetic differences,13different traits associated and its heterosis relations were analyzed. At the same time, the Oligo (dT) method made the innovation in the reverse transcriptase cDNA process. The main results are as follows:The SRAP molecular marker results of hybrid corn in the original parent through cDNA level show that there certainly were genetic differences that expression between corn hybrid and their parents. It can be divided into6types, named single express consistent type (110type), single express consistent type2(011type), parents were silent type (101type), single express silent type1(100type), single express silent type2(001type) and hybrid specifically expressed type (010type).The study about6kinds of differences expression pattern between hybrid and parents found that parents were silent type, single express silent type1and type2, this three kinds of differences expression patterns accounted for46.13%of the sum total of the expression, the results of this study can explain that parent genes express silence is the main form in the hybrid progeny of genetic differences. Hybrid specifically expressed in this trial type also had the high frequency, accounting for14.9%of the total express all differences, because of individual genes interactions or environmental interactions caused a mechanism of produce offspring or activate in their representatives in the hybrid progeny, thus showing specificity.Do the related analysis about six different differential expression patterns and characters of13heterosis, the results showed that in the15correlation coefficient of differences expression and heterosis,10happened in agronomic traits, only five happened in quality traits. This shows that the heterosis of corn grain represent the future generations in the field of the seed plants, different types of differences in the expression pattern also get intuition genetic expression, the three agronomic characters about plant height, grain number per line and fresh weight were very significant relationship in different expression mode. Among them, fresh weight per is the only one with the hybrid specifically expressed type (type010) presented related, and for very significant related characters. It indicated that this type for corn yield traits of heterosis had a positive effect. In the reverse transcription cDNA method, the study of the creative used the method combine random primers with the primer Oligo (dT), that which was in the synthesis of the first chain was also join cDNA random primers and Oligo (dT) primer, this is not only ensure the advantages of random primers big production, but also used the advantages of the specificity strong of Oligo (dT) primer. Such methods has not seen any reportsat at present, it is the first application in this lab. The random primers in our method and the new method were compared in this experiments, and it was obvious that the new method of synthesis of cDNA strip were more clearer than first chain, the content of cDNA were more higher.
