In north China beet is an important economic crop, it is the world’s most important sugar crops. But there is many diseases in the production of sugar beet, and its yield and quality is affected badly. Beet necrotic yellow vein virus infecting Sugarbeet after plexus root symptoms (Rhizomania), and led the yield and quality of sugar beet sharp decline.In many methods of sugar beet disease control, breeding of fine resistant cultivars is the most effective means. Ribosome inactivating protein (RIPs) is a kind of protein on virus, fungi, bacteria, resistant class can inhibit bacteria growth and proliferation ability.This experiment in our task group adopts PCR.RT-PCR to identify the first and second generation of transgenic sugar beet which turn RIPs gene. And then we do the research to Western blot、form、yield and quality identification and physiological nature. From the first generation and the second generation of RIPs transgenic beet, screen for genetic stability, strong disease resistance in transgenic plants, for sugar beet breeding for disease resistance in selected base material, to provide a reliable research materials. The results of the experiment are as follows:1Detection and Screening the transgenic sugarbeet which has target gene by PCR. The RIPs gene expression of the transgenic sugarbeet which have been detected by RT-PCR,showed that RIPs gene had integrated into the genomic DNA of sugar beet and expressed at the level of transcription. This experiment provide reliable materials for breeding the sugar beet with the disease resistance.2The RIPs transgenic beet is first planted in the BNYVV soil, with wild beet as controls, The results show that, in diseased-free soil RIPs transgenic sugarbeet mean weight lower than the wild type sugar beet, average sugar content higher than the wild type sugar beet; in the diseased soil, RIPs transgenic beet average weight is higher than the wild type, the average percentage of sugar than wild type lower.3Western blot the results showed that the RIPs gene transferred to Sugar beet made the resistance to BNYVV disease has greatly improved.4photosynthesis, nitrogen metabolism, defense enzyme, resistance index of the transgenic sugarbeet which have been screened, The research showed that contain RIPs transgenic sugarbeet chloroplast content, soluble sugar content, SPAD value, chitinase and β-1,3-glucanase activity and polyphenol oxidase activity is higher than wild-type plants, and transgenic plants against pathogens have higher inhibition, resistance is stronger.
