Cut rose flowers (Rosa hybrida) have a great of prospects and consumer demand in the flower market. But the cut rose flowers are always under water stress due to the harvesting, processing, transportation and other factors. Water stress accelerated senescence process of cut rose and further affected the life and quality of cut flowers. So improving the tolerance of cut rose to water deficit is very useful and practicability. Zhao Xiting et al. have made a research about endopeptidase in cut rose petals under water deficit and the research showed that water deficit significantly induced the serine protease activity. My experiment on the basis of cloned serine protease RhSepl in the cut rose petals under water deficit was to explore the role in the process of dehydration stress.To better follow-up study, at first I made a bioinformatics analysis to RhSepl gene, which is a comprehensive understanding of the RhSepl gene, including the molecular weight; transmembrane; hydrophobicity; signal peptide; homology, functional sites and other physical and chemical structure. All of these findings were foreshadowing of the next research of the cut rose serine protease RhSepl in the dehydration stress. The obtained RhSepl gene recombines Pet-28a (+) vector to construct the recombinant expression vector called Pet-28a-RhSepl. And Pet-28a-RhSepl was transfected into E. coli, which was induced to express serine protease. SDS-PAGE and Western blot results showed that the serine protease degrades seriously. Expression optimization and other bacteria express system do not make the target protein expressed without degradation. RhSepl gene relative quantitative PCR showed that the expression of RhSepl gene increased with water deficit stress intensity, but the protease is not by dehydration stress-induced produced. The contents of this experimental study suggest the cloned cut rose petals protease RhSep1involved in the rose dehydration stress physiology. The biochemical properties of the RhSepl and in the process of dehydration stress both need to further study.
