ObjectiveThe present study was conducted to investigate the RhCMV infection rates and make the PCR identification for RhCMV for rhesus monkey from the National Experiment station (Ya’an City, Sichuan Province). Histological distribution of RhCMV from main organs and the hypothalamus for adult and embryonic rhesus monkeys, were systematically detected to find the relationships between the distribution of RhCMV and low immunity, induced cell apomorphosis and abortion, which were served as an effective basis on the research of RhCMV and HCMV and other wildlife protection in the future and provided a reference on the quarantine standards of the valuable animal model for rhesus monkey.Method(1) The RhCMV-IgG antibody of the blood was qualitatively and quantitatively detected by using the elisa kit of RhCMV-IgG antibodyb for100adult female rhesus monkeys(2) Primers design was conducted to make PCR identification with RhCMV from the lymph nodes for rhesus monkey(3) Probes design and ISH were used to making histological detection of RhCMV from main organs (heart, liver, spleen, thymus, trachea, lungs, kidneys, intestines, stomach,umbilical cord, placenta, uterus, testis,ovarian) and the hypothalamus for adult and embryonic rhesus monkey.Results(1) The qualitative and quantitative infection rates of RhCMV were76%and73%for100adult female rhesus monkeys, respectively. The antibody concentration was mostly from10to30pg/ml. These results indicated that infection rate of RhCMV was more than70%because of the similarity between the qualitative infection rates and the quantitative infection rates.(2) The partial RhCMV gB genes were Successfully amplificated by the primer design that was based on RhCMV gB gene and the template of which was from DNA extract for rhesus monkey lymph node tissue, which concluded that there was higher probability of RhCMV infection rates for experimental rhesus monkey.(3) Histological detection of RhCMV was made by the method based on methybridization in situ for adult and embryonic rhesus monkeys, the findings indicated that there were quite difference in different organs color and the positive product sizes for testing slices, which were almostly distributed with blue-blt ack granular or punctate. The viruses were distributed in nucleus, cytoplasm, membrane and Stroma of cell, However, virus-positive signals were relatively inferior. Histological distribution of RhCMV was similar between adult and embryonic rhesus monkeys. There have been no blue-black positive products in the negative control group and blank control group for rhesus monkeys.Viruses were detected in the organization of infected rhesus monkeys by in situ hybridization probe for RhCMV gB gene. The main distribution sites inclueded as follows: The heart of theendocardium, The nuclei and Stroma of liver cell in rhesus monkey fotus,The lymphocytes of red pulp, white pulp and marginal zone of the spleen; The lymphocytes of the cortex, medulla and the junction between cortex and medullaat in thymus; The mucous layer and submucosa of the trachea; The Ⅰ alveolar cells and Ⅱ alveolar cells in Breathing Position of lung; The renal capsule, renal tubule and Renal corpuscles; The Mucosa, intestinal villi, lamina propria and muscularis mucosa of intestine; The mucous layer of the stomach; The connective tissue of umbilical cord; The fetal dense chorionic and maternal base decidua of Placenta; The myometrium and endometrium; The myoid cells of seminiferous tubules and Mesenchymal cells in Testicular; The granular layer of the follicle in Ovarian cortex, and ovarian sarcolemma; Periventricular nucleus, the preoptic medial nucleus of thepreoptic lateral nucleus, anterior nucleus, paraventricular nucleus,supraoptic nucleus, dorsomedial nucleus, dorsal lateral nucleus,ventral lateral nucleus, ventromedial nucleus, optic tract, arcuate nucleus, central gray and so of the hypothalamus.In summary, there were viruses distributed in most organs for rhesus monkey, especially quite a few in the immune organs, reproductive organs, and hypothalamic organs. Based these results, we strongly suggested that RhCMV may be the main reason of resulting in different impact on all organs of the body, and lead to lower immunity, reduced fertility, nervous system dysfunction and other symptoms for rhesus monkey.
