Vasoactive intestinal peptide is composed by28amino acid alkaline peptide, it is a member of secretin hormone-glucagon family. VIP and its receptors are widely distributed in all organs of animal body, and mediate various biological activities. In recent years, VIP had been studied mostly in the field of immunization and cancer treatment. There are few reports about specific effect and result of regulation on body all stages of development. So this study proved that the distribution of VIP and its mRNA in the ovary and endometrium of mouse at the stage of puberty, pregnancy, lactation period and late lactation.The research about VIP will be provided as the morphological basis for researching in regulation of the reproductive system in future.Using immunohistochemiscal PV-9000two-step and in situ hybridization methods to detect the distribution of VIP and its mRNA in the ovary and endometrium of puberty, pregnancy, lactation period and late lactation of mouse. The results showed that VIP-positive cells were mainly expressed in luteal cells and oocytes in mouse ovary. They mainly located in parts of the cytoplasm of luteal cells, no located in the nucleus and cell membranes. The other positive parts were in thecal and oocyte membrane of primary follicles, cytoplasm and nuclei did not express immunoreactivition The three periods also showed immunoreactivity in cytoplasm of granulosa cell except puberty. By single factor analysis of variance showed that in the mouse ovary, VIP immunoreactive substance was mainly expressed in the luteal cells and follicles,and expression of late lactation was significantly different (P<0.01)compared to other three stages. The VIP-positive cells are also mainly expressed in endometrial epithelium cell, cytoplasm and nuclei did not express immunoreactivition. The endometrial glandular epithelial cells were positive Immunoreactivity, the nucleus not had Immunoreactivity substance. The stromal cells were positive Immunoreactivity, cell membrane and cytoplasm had Immunoreactivity substance. By analysis of variance showed that in the mouse endometrial, the expression of uterine glandular epithelium cell at the pregnancy were signi ficantly different (P<0.01) compared to other three stages.It made the conclusion,VIP and its mRNA expressed in the cytoplasm of luteal, internal thecal of follicule and oocyte membrane in the ovary.They ere expressed in the cytoplasm of granulosa cells except puberty. In the four physiological stages of mouse, VIP and its mRNA were expressed in the same parts of ovary, and their expression levels increased with the increase of age. The maximum expression amount was found in the late lactation of mouse. The VIP-positive cells are also mainly expressed in endometrial epithelium cell, uterine glandular epithelium cell and stromal cell in endometrial of four stages mouse, and cell membrane and cytoplasmic were stained. The maximum expression amount was oberved at the stage of pregnancy, and was significantly higher than that of the other three stages.
