Comparison of some immune traits and bacterial burden, mRNA expression of key genes involved in NLRC5signaling pathway were investigated between young healthy birds and infected chickens with salmonella(SE) and Poly I:C in Langshan and Ross breeds,aiming to investigate functional genetic markers associated with disease resistance breeding and more of molecular mechanism for immune response of SE infection and Poly I:C.1. For the chickens of Langshan, the serum concentrations of IgY and IFN-y of group1,2, and3were significantly higher than that of control group;the serum concentrations of IgM of group2was significantly higher than that of control group; the serum concentrations of IL-1β of group1was significantly higher than that of control group. For Ross chickens, the serum concentrations of IgY of group1and3were significantly higher than that of control group;the serum concentrations of IgM of group1,2, and3was significantly higher than that of control group; the serum concentrations of IL-1β of group2was significantly higher than that of control group.2. The results of gene expressions in Langshan chicken tissues infected with Salmonella enteritidis were as follows:the mRNA expression level of TLR4, TRAF6, TAB and NLRC5genes in thymus of group1was significantly higher than that of control group; the expression level of NLRC5gene in ceacum of group1was significantly higher than that of control group; the expression level of NLRC5, IKKβ and IKKa genes in liver of group1was significantly higher than that of control group; after infected with Salmonella pullorum, the mRNA expression level of TAK1, NLRC5and NF-κB genes in thymus of group1was significantly higher than that of control group;the expression level of IKKβ gene in spleen of group1was significantly higher than that of control group;the expression level of IKKβ and NF-κB genes in ceacum of group1was significantly higher than that of control group, the expression level of NF-κB and IKKa genes in liver of group1was significantly higher than that of control group; after infected with Poly I:C, the mRNA expression level of TRAF6, TAB and NF-κB genes in thymus of group1was significantly higher than that of control group, the expression level of TAK1gene in spleen of group1was significantly higher than that of control group; the expression level of TLR4and TAB genes in ceacum of group1was significantly higher than that of control group; IKKa gene in liver of group1was significantly higher than that of control group.3. The results of gene expressions in Ross tissues infected with Salmonella enteritidis were as follows:There was no significant difference of the mRNA expression of all genes in immune tissues between group1and control group;the expression level of IRAK4and NF-κB genes in ceacum of group1was significantly lower than that of control group;the expression level of IKKa, IRAK4, MyD88, NF-κB and NLRC5genes in liver of group1was significantly lower than that of control group; after infected with Salmonella pullorum, there was no significant difference of the mRNA expression of all genes in immune tissues between group1and control group, the expression level of IKKa、IRAK4, MyD88, NLRC5, TAB and TLR4genes in liver of group1was significantly lower than that of control group; after infected with Poly I:C, there was no significant difference of the mRNA expression of all genes in immune tissues between group1and control group;IRAK4gene in ceacum expressed significantly higher in group1than in control group, and IKKa and NLRC5genes in liver expressed significantly higher in group1than in control group.4. After infected with Salmonella enteritidis, Salmonella pullorum and Poly I:C, the key genes involved in NLRC5signaling pathway, presented significant expression in immune tissues in Langshan chicken, while no significant expressions were detected in Ross chicken. The reason may becaused by the differences of genetic background andthe innate immune performance between Langshan and Rosschicken.
