Avermectin (AVM) was a large ring lactone disaccharides compound which used as insecticideand acaricide for animal and agriculture. It had an excellent helminthic effect and security. But withthe increasing use of AVM, there were more and more accumulation in the environment. And it hada bad effect on ecological system. The pigeon had a very strong sensitivity of the chemical changesin the environment. The liver and kidney of king pigeon in vivo were the research targets which fedwith AVM at final concentrations of0mg/(kg·diet),20mg/(kg·diet),40mg/(kg·diet) and60mg/(kg·diet) respectively in30d,60d and90d. And chronic AVM poisoning model wasconstructed. Pathologic histology, changes of ultrastructure, oxidative stress, DNA injury,cytochrome P450(CYP450) enzyme system, apoptosis, caspase-3, caspase-8and fas mRNAexpression level were detected to study toxicity mechanism in liver and kidney caused by AVMrespectively in30d,60d and90d. It would provide the scientific evidence for overall assessmentof the ecotoxicology effect of AVM on birds. The results showed as followed:1. HE staining, Transmission electron microscopy were selected to observe the toxicity ofAVM at different dosages and times in liver and kidney. The results showed that AVM couldinfluence the normal structure in liver and kidney observed from morphology.2. By detecting the activity of anti-oxidative index, the content of carbonyl and DPC, theresults revealed that AVM could induce oxidative stress of liver and kidney, decrease activities ofGSH-Px and SOD, increase the contents of MDA, carbonyl and DPC rates in different groups. Ithinted that AVM could lead to oxidative stress of king pigeon’s liver and kidney and would affectits normal physiological function, further induce the apoptosis.3. The apoptosis and caspase-3, caspase-8, fas mRNA expression level in liver and kidneywere detected by fluorescent quantitation RT-PCR and TUNEL assay,the results showed that AVMcould induce apoptosis and increase caspase-3, caspase-8, fas mRNA expression level in pigeonliver and kidney in a concentration-dependent manner, which were the signature genes of apoptosis.4. By the detection of the contents and activity of CYP450enzyme system, it showed thatAVM could decrease the contents of CYP450and b5, and the activity of CR, AND, ERND, AH,which proved that both CYP450enzyme system and oxidative stress of organism participated in thedamage of AVM on the liver and kidney. In a word, AVM could affect the function of cytochrome P450enzyme system and oxidationstress which cause issue morphology, and then induce apoptosis by expression of caspase-3,caspase-8, fas mRNA in liver and kidney. It provided information for further research and provideda scientific basis to protect ecological environment and ensure the development of graziery.
