Effects Of Traditional Chinese Medicine Extract The Proliferation And The Expression Of Related Protein Schwann Cells

For studying the effects of Frankincense Extract, Codonopsis pilosula extracts, andAngelica extracts the indications including the safe medication range, the cell growth curve,the phase change of cell cycle, the content of BDNF, NCAM and PCNA were in liversupernatants of SCs. It was better to research the mechanism of improving the wound healingof nerve repairing and regenerating.Schwann cell was chosed as the experimental object. The cell toxicity and cell growtheffects of ME, CPE and AE was detected by using MTT method. The cell cycle of Schwanncell was detected by using FCM. The sCPEetions of BDNF, NCAM and PCNA were detectedby using ELISA kits. ELISA kit was used to detect the content of BDNF, NCAM and PCNA.Time effect of concentration of traditional Chinese medicine extract on the cytokine BDNFmRNA of SCs by RT-PCR.The results showed that:(1) The safe medication range of ME, CPE and AE effecting onSchwann cells were respectively62.5~8000mg/L、125～4000mg/L、125～2000mg/L.During the Chinese herbal extract intervened Schwann cell within24and48hours, theresults of MTT by traditional Chinese medicine showed that different concentration of ME,CPE and AE were respectively125mg/L～500mg/L,1000～4000mg/L and500～2000mg/Lto promote cell proliferation and the control group (P<0.05).(2) The results of flowcytometry method during the Chinese herbal extract intervened Schwann cell within24and48hours showed that they promoted cell cycle and the percent content of DNA by theconcentration of ME were125、250and500mg/L; the concentration of CPE were1000、2000and4000mg/L; the concentration of AE were500、1000and2000mg/L and the controlgroup (P<0.05).(3) ELISA kit was used to detect the content of BDNF, NCAM andPCNA.The results showed that all have the different of traditional Chinese medicine toincrease.(4) The results of RT-PCR showed that different concentration of traditionalChinese medicine on the expresstion BDNF mRNA of SCs to increase, and ME, CPE andAE could promote the expresstion of the BDNF mRNA at concentration of125,2000and 1000mg/L, respectively.Conclusion: The extract at certain concentration was effective on inducing theproliferation of SCs. and the best concentration of MA, CPE and AE were125,2000and1000mg/L.At the same time, they could up-regulate the expression of BDNF, NCAM andPCNA from SCs cultured in vitro.
