Soil microbial secondary metabolites have always been a major source of active leadcompounds, and in order to find more active lead compounds from soil microorganisms, the activeStreptomces sp.NEAU-Z4was isolated from the soil in our laboratory. In order to detect andanalyze the secondary metabolites of the strain, in this article the secondary metabolites of thestrains were studied carefully as following:(1) Streptomces sp.NEAU-Z4was fermented with the volume of50liters, fermentationmedium was made of0.4%yeast extract,4%soluble starch,1%glucose,1%soybean meal,0.1%NaCl,0.2%K2HPO4,0.1%MgSO47H2O and0.2%CaCO3with the pH value of7.2-7.4. Afterconfigured with distilled water, the fermentation medium was sterilized for20minutes at121°C.With the inoculum size of8%, the seed liquid was sent to50liter fermentor and cultured underconditions, including28°C, a stirring rate of100r/min and a ventilation rate of120m3/h, for6-7days.(2) The mycelia cake was obtained after the fermentation broth filtrated through the filtercloth, then washed with acetone soak overnight. The filtrate was adsorbed onto a Diaion HP-20column and the column was eluted with industrial ethanol filtrate. After detected by TLC, theacetone soaking and ethanol eluent were combined together and concentrated under reducedpressure conditions, and then extracted three times with an equal volume of EtOAc. As a result,10compounds were isolated from the extract with the methods of silica gel column chromatography,Sephadex LH-20gel column chromatography, preparative HPLC chromatography and semi-preparative HPLC chromatography.(3) All the compounds were characterized by the spectral analysis methods of1H-NMR,13C-NMR, IR, UV, mass spectrometry,1H-1H COSY, HMQC, and HMBC. At last, we found that8compounds are novel compounds,5-But-2-enylidene-1,2,3,5-tetrahydro-[1]pyrindin-4-one (1),Dodeca-8,10-diene-1,5-diol (2),1,3-Dihydroxy-dodeca-8,10-dien-5-one (3),5-But-2-enylidene-1,5-dihydro-[1]pyrindin-4-one (4),Dodeca-8,10-diene-1,3,5-triol (5),1-Hydroxy-dodeca-3,8,10-trien-5-one (6),2-Hepta-3,5-dienyl-2-methoxy-tetrahydro-pyran-4-ol (7),1-(2-Hepta-3,5-dienyl-2-methoxy-tetrahydro-pyran-4-yloxy)-3-hydroxy-dodeca-8,10-dien-5-one (8). The remaining2areknown compounds,6-(3’-methylbuten-2’-yl) isatin (9) and1H-indole-3-carboxylic acid (10). Thecompound1express activity against human prostate cancer PC-3with IC5054.7μg/ml. |
