Weeds can reduce farm income by up to30%and the cost of developing new herbicides is20–100times higher than the cost of generating new varieties of herbicide-resistant crops.Therefore, Herbicide-resistant crops could effectively control weeds, increase laber productivity,decrease planting cost and protect crops from herbicide injury which had important economicvalues.Sreening and identification were different wild soybean resistace to glyphosate in thisresearch using field trials. Wild soybeans were to study the stability and resistant extent of them.Target enzymes, protect enzymes and endogenous hormone have finished for analysis, thusphysiological and biochemical mechanism to glyphosate is clear. The expression and function ofglyphosate-resistant EPSPS gene provided the theoretical basis for herbicide-resistant mechanism.Cloning resistance gene and breeding are importment significance of herbicide-resistant cropresistance to glyphosate in the further. The main findings of this study as follows:1. The92wild soybean materials showed significantly different resistance levels after theapplication of1230g ae ha-1glyphosate application and acquire the two resistant wild soybeangermplasm (ZYD254and ZYD0685). At different dosages and different years, ZYD254is stablestrain and high temperature has some effect.2. The resistant material ZYD254and susceptible materials ZYD1158the shikimateaccumulation and chlorophyll content were analyzed. At glyphosate application, there were nosignificant differences in shikimate levels and Chlorophyll content in ZYD254. But there havesignificant decrease in shikimate and chorophyII content in ZYD1158. Therefore, shikimatecontent no accumulation was a reason for physiological mechanism.3. Resistant wild soybean leaves after glyphosate stress, the leaves of SOD, POD showeddecreased and then gradually,MDA content increased and then decreased. But ZYD1158continued to decrease. Therefore, Resistance strains were higher activity cleared reactive oxygenmade of wild soybean. This factor is one of the reasons for the enhancement of the resistance ofwild soybean GSTs and GSH activity was enhanced in ZYD254.4. The resistant strains endogenous hormones can restore a short period of time at glyphosatestress. IAA and GA content were higher increase in ZYD254. ABA content is slightly lower. IAAand GA content were decreased, ABA content constantly increased in ZYD1158. 5. By bioinformatics analysis showed that wild soybeans in two GmEPSPS gene copies, Inthe current results EPSPS1mRNA levels were increased about2-4fold in ZYD-254and peak levels were observed at6h after treatment with1230g ae ha-1glyphosate.
