| Acer rubrum’Brandywine’, which belongs to the family of Acer, is a high deciduous arbors and originated in the United States. Because of its strong adaptability, temperature resistance, drought tolerance and higher growth vigor, Acer rubrum is one of the most excellent species in landscaping. In order to solve the supply of the Acer rubrum seeding, it is an effective way to carry on tissue culture of Acer rubrum.This experiment adopted the leaf, the stem segment and the axillary bud as explants, focus on researching the effects of basic medium, growth hormone and other additives on the tissue culture of Acer rubrum in a direct and indirect organogenesis way systematically. Through above researches, the suitable medium in each stage have been found out, and get Acer rubrum regenerated plants successfully, established Acer rubrum plant regeneration system. The main results are as follows:(1) The difficult degree of sterilization was different for various explants, as well as the same explants in each season. As to different explants or sampling time, the best explants is stem segment, and suitable sampling time is from March to May, which the contamination rate is only5.4%and the survival rate is88.5%.(2) In initiation culture, the budding rate higher when the culture medium is WPM, the IBA concentration0.5mg/L and the6-BA concentration2.0mg/L. So the suitable initiation culture medium for Acer rubrum is WPM+IBA0.5mg/L+6-BA2.0mg/L.(3) Five factors’influence order to callus induction is medium> explants>NAA> KT>6-BA. The Acer rubrum induction rate is reached the highest, when cultured in the WPM, stem segment as explant, the NAA concentration0.7mg/L, the KT concentration0.6mg/L or the6-BA concentration0.4mg/L. Thus, WPM+NAA0.7mg/L+KT0.6mg/L+6-BA0.4mg/L and the stem segment as explant was more appropriate for the Acer rubrum callus induction.(4) When Acer rubrum callus cultured in medium with various hormone at each level, there was only the increase of the callus mass, and the colour was turned from pale yellow to light or dark green, but no bud formation. What’s more, the quality of Acer rubrum callus decreased constantly as the frequency of subculture increasing.(5) Culture medium and hormone concentration plays an important role in the root induction. The range analysis displayed that four factors’influence order is6-BA>GA3>medium>IBA. MS+IBA2.0mg/L+6-BA0.0mg/L+GA31.0mg/L was the most suitable combination for root induction, which rooting rate reached93.3%. It is still the ideal combination according to the k value of the four factors. Secondly, the suitable combination is WPM+IBA1.5mg/L+GA30.5mg/L, which rooting rate was80.0%.(6) Different methods of training seedling have obvious effect on the survival rate of tissue culture seedling, before transplant. Place the seedling6days without opening the bottle, and then open the bottle for2days, the average survival rate reaches the top of86.7%.. Another option is place the seedling6days without opening the bottle, and then open the bottle for3days, on which the average survival rate is83.3%. Different transplanting mediums have significant effect on the survival rate of Acer rubrum seedlings. The favorable transplanting substrate is the sand and humus soil, which should be mixed equally, which lead a high average survival rate81.1%. |
