Paclitaxel is widely used for treatment of cancer. However, paclitaxel is limited by its Water-insolution and short half life. The aim of this study was to develop injectable sustained-release formulation. The dosage form constituted by biodegradable polylactide and sustain release of paclitaxel in the tumor.The PLA drug-loaded microspheres were prepared by solvent evaporation. Investigated the surface morphology, particle size distribution, drug loading rate and encapsulation efficiency and in vitro drug release behavior of microspheres. The results show that the drug-loaded microspheres had a spherical shape, uniform particle size distribution, high rate of drug-loading encapsulation, microsphere stability.After sterilized byγ-irradiation, the biological safety of the polymer was evaluated by sensitization, skin irritation, hemolysis and bone marrow micronucleua teat. The results show that the PLA drug-loaded microspheres were biocompatibility and low toxicity, which can be used as Injectable sustained-release formulations.Examine the retention organ, retention time and liver toxicity of the PLA drug-loaded nanoparticles. The results showed that the drug-loaded nanoparticles in rats mainly retented by the liver, and metabolites for 4 days. Watch the rat liver slices under electron microscopy, The results showed that the drug-loaded nanoparticles can nt affect the liver cells. Examing the five indicators of liver function of the rat which were of injected drug-loaded nanoparticles. There was no significant change compared with normal rats in the five indicators of liver function。...
