Objects:This paper is discussed about the influence of plant estrogen alpha zeranol and estrogenreceptor antagonist to rat osteoblasts secrete cells factors, Study of the mechanism of plantestrogen alpha zeranol on postmenopausal osteoporosis prevention and cure function.Methods:1、Add the different concentration of α-ZAL (10~-5mol/L~10~-10mol/L) in osteoblastculture system, use Elisa and half quantitative RT-PCR method to detect the secretion changesand the influence of the mRNA expression of OPG, IL-6, TNF-alpha of rats osteoblast,respectively in24h,48h,72h.2、Add the α-ZAL (10~-6mol/L) and different concentration of estrogen receptorantagonists(10~-5mol/L,10~-7mol/L,10~-9mol/L)in osteoblast culture system, use Elisaand half quantitative RT-PCR method to detect the secretion changes of OPG, IL-6,TNF-alpha of rats osteoblast respectively in24h,48h,72h,also detect the influence of themRNA expression of OPG in72h.Result:1、The results found that the different concentration of α-ZAL(10~-5mol/L~10~-10mol/L)act on osteoblasts, along with the increase of the concentration of α-ZAL and the extension ofeffect time, the secretion capacity and mRNA expression of the OPG have a different degreeof increase, but IL-6, TNF-alpha have a different degree of reduced.α-ZAL can promote theosteoblast to secrete and express mRNA of the OPG, restrain IL-6, TNF-alpha secretion andIL-6mRNA expression.2、The results showed that α-ZAL(10~-6mol/L) and different concentration of estrogenreceptor antagonists(10~-5mol/L,10~-7mol/L,10~-9mol/L)act on osteoblasts, when α-ZALseparate function, the secretion and mRNA expression of the OPG are increased, the secretioncapacity of IL-6, TNF-alpha are reduced; When α-ZAL in company with estrogen receptorantagonists function, the secretion and mRNA expression of OPG have different rate ofreduction, the secretion capacity of IL-6, TNF-alpha have different rate of increases.Conclusion: 1、α-ZAL has a similar effect like estrogen, which can stimulate the osteoblasts to releaseOPG, restrain the broken bone cells to release the differentiation of cytokines IL-6,TNF-alpha, thereby it can reduce the vitality of broken bone cells, inhibit the differentiationand mature of the broken bone cells, which lead to enhance the bone formation ability ofosteoblasts and decrease the absorption ability of broken bone cells.2、The above results suggest that the osteoblasts express OPG, IL-6, TNF-alpha can beblocked by estrogen receptor antagonist, which suggests the role of α-ZAL is mediatedthrough estrogen receptor....
