Objective To analysis the frequency distribution of Insertion/Deletion (I/D) polymorphism in theACE, Discusse the association between pulmonary artery pressure, blood oxygen partial pressurewith the Insertion/Deletion (I/D) polymorphism frequency distribution in the ACE, Reveal therelationship of Adaptation to High Altitude in Tibetan. The research from the genetic level furtherlyexplored genetic and environmental factors to human impact.Methods To select30Tibetan young men,19-21years old who live in area of3430±333.96meters, whose parents are all Tibetan and haven’t intermarry with other nation among threegenerations, as the experimental group.Selected the same conditions of the30Han chinese youngmen as control group. The I/D polymorphism of ACE gene are detected by PCR. The two nurses willcollect2ml radial artery blood by BD Preset vessel, immediately,professionals analyse bloodoxygen partial pressure value with AVL instrument; Color doppler ultrasound analyzer is used tomeasure pulmonary artery pressure, to caculate pulmonary artery pressure value by the simplifiedBernoulli (Bernoulli) equation. Elisa test detected the ACE level in the plasma. To get ACEconcentration through the drawing standard curve.Results The PCR amplification show that the NO.16introne of ACE gene augmented can formtwo different length segment of DNA, that is490bp Insert segments and190bp and Desert segments.The product after dyeing, electrophoresis results are shown under ultraviolet lamp that there arethree bands: one of them is a single strip of the490bp for the homozygous form the insertion type(Ⅱ); one of them is a single strip of the190bp for the homozygous form for desert type (DD); one ofthem are two strips of the190bp and490bp for the heterozygous form for the insertion type(Ⅱ)and the desert type (DD). The three genotypes II、ID and DD of ACE were detected both in theexperimental group and the control group, they were II、ID and DD. The frequency of the threegenotype was0.700、0.233、0.067in the study group, while the control group were0.300、0.467、0.233. The genotype of ACE was significantly different between the two group(χ~2=9.725,p=0.006);The I/D allele frequency were0.817,0.183and0.534、0.466,between the two groupwhich Was significantly different(χ~2=10.227,p=0.001). Differences in the two groups ofpulmonary artery pressure were not significantly differen(tp=0.605,p﹥0.05); Differences in the twogroups of Artery blood oxygen partial pressure were not significantly different(p﹥0.05). Thedifference of plasma ACE have statistically significant (p﹤0.05).Conclusions The insertion/deletion (I/D) polymorphism of ACE gene have been different in the Tibetan and Han Chinese. the overrepresented I allele of the ACE gene in Tibetan might be one offundamental geneic factors responsible for adapting to a high-altitude environment.
