[Objective]To study the changes of expression of proliferating cell nuclear antigen (PCNA),p27and PTEN in normal oral mucosa,oral leukoplakia and oral squamous cellcarcinoma of human and normal oral mucosa,hyperplasia tissue and oral squamouscell carcinoma of rat by using two-step method of immunohitoch-emistry.And toinvestigate the role of PCNA,p27and PTEN in the process of oral normal mucosa,oral precancerous lesion and oral squamous cell carcinoma,and then compare theexpression mentioned above with the expression of PCNA and p27in ratcarcinogenesis model.The study and investigation can provide molecular evidence fordetecting oral leukoplakia carcinogenesis in clinical practice and offer reference in theclinical application of anticarcinogenic medicine.[Method]1.To apply two-steps method of immunohistochemistry to detect the expression ofPCNA,p27,PTEN in10cases of normal oral mucosa,15cases of spot not withhyperplasia leukoplakia,10cases of spot with hyperplasia leukoplakia and20cases ofsquamous cell carcinoma.Statistical software is applied to analyze the experimentalresults.2.Apply two-steps method of immunohistochemistry to detect the expression ofPCNA and p27in10cases of normal oral mucosa,10cases of spot not withhyperplasia leukoplakia,15cases of spot with hyperplasia leukoplakia,10cases ofsquamous cell carcinoma from the rat carcinogenesis model.Statistical software isapplied to analyze the experimental results.[Result]1.The expression of PCNA in normal oral mucosa,spot not with hyperplasialeukoplakia,spot not with hyperplasia leukoplakia and squamous cell carcinoma ofhuman is gradually increasing.The statistical result shows that the expressiondifference of PCNA is of statistical significance in normal oral mucosa and spot notwith hyperplasia leukoplakia,compared with spot with hyperplasia leukoplakia andsquamous cell carcinoma (P<0.05). 2.The expression of p27and PTEN in normal oral mucosa,spot not withhyperplasia leukoplakia, spot with hyperplasia leukoplakia and squamous cellcarcinoma of human is gradually decreasing.The statistical result shows that theexpression difference of p27and PTEN is statistically significant in normal oralmucosa and spot not with hyperplasia leukoplakia, compared with spot withhyperplasia leukoplakia and squamous cell carcinoma (P<0.05).3.The correlation of PCNA,p27and PTEN:The result of Spearman statistical testshows that the expression of PCNA is negatively correlated with PTEN and p27inspot with hyperplasia leukoplakia and squamous cell carcinoma of human,but that ofPTEN is positively correlated with p27.4.During the carcinogenesis process of normal oral mucosa,spot with hyperplasiamucosa and squamous cell carcinoma of rat,the expression of PCNA tends toincrease,and that of p27tends to decrease.Therefore,the expression differences ofPCNA and p27in these three tissues are both of statistical significance.The expressionof PCNA is negatively correlated with p27in abnormal hyperplasia tissue.Theexpression of PCNA is negatively correlated with p27in squamous cell carcinomatissue.[Conclusion]1.The expression of PCNA in the carcinogenesis process from oral spot not withhyperplasia leukoplakia to oral squamous cell carcinoma trends to increase,but theexpression of p27and PTEN trends to decrease.It demonstrates that positiveregulation of PCNA is motivated during process of oral precancerous lesion and oralcarcinoma and accelerates the cell proliferation.However,the negative regulation ofp27and PTEN is restrained or lost,and cannot suppress the cell proliferation.Andthis promotes the formation of tumor.2.The expression of PCNA and p27in normal oral mucosa,spot with hyperplasialeukoplakia and squamous cell carcinoma of human is similar to that in rat oralcarcinogenesis model.3.PCNA and p27reflect the histological changes in the process of oral carcinogenesis properly.They can be considered as “marker molecule” for detectingoral precancerous lesion in clinical practice.And the changes of expression canprovide clinical basis for molecular targeted therapy....
