【Objective】To investigate the relationship between pathological and the expression ofClaudin-1or Claudin-5and clinical parameters in normal tissue、colorectal adenomaand colorectal carcinom.The change of Claudin-1and Claudin-5in the developmentof colorectal cancer occurrence is studied, and their relevance is analyzed to providethe basis for carrying out the diagnosis and treatment of colorectal cancer gene.【Methodes】Using complete clinical and pathological data, Yan’an University AffiliatedHospital2008-2011surgical resection specimens, all patients had not receivedradiotherapy, chemotherapy or other tumor-related treatment. The expression ofclaudin-1and claudin-5in the application of immunohistochemistry in the SP(Streptavidin Perdcidase) detection of15pathologically proven cases of normalcolorectal mucosa (normal group),15patients with colorectal adenoma (adenomagroup),17patients without lymph node metastasis and23cases with lymph nodemetastasis in colorectal cancer (carcinoma group).【Results】1.The positive rate of Claudin-1expression in normal, adenoma, carcinomagroup were33.33%,46.66%,75.00%,those suggests that multiple groupsdifferences among the groups were statistically significant(p﹤0.05)。Claudin-1expression in poorly differentiated positive expression rate was90.00%, positiveexpression of the differentiated group was56.00%, the score group, positive expression rate was20.00%--there was significant difference between groups (P<0.05);In Dukes staging A+B, the positive expression rate was60.00%,and thepositive expression rate was92.00%in C+D, the two groups had significantdifference(P<0.05); The positive expression of the lymph node metastasis groupwas86.95%higher than those without lymph node metastasis positive expressionrate, which was52.94%, the difference was significant (P<0.05). The positive rate ofClaudin-1expression is not correlated with gender, age and tumor size.2. Claudin-5positive expression rate in the normal group, adenomas, cancergroup were86.66%、53.33%、45.00%suggesting that the difference between thegroups was statistically significant (p>0.05);Claudin-5expression in poorlydifferentiated positive expression rate was40.00%, positive expression of thedifferentiated group was88.00%, the score group, positive expression rate was90.00%--there was significant difference between groups (P <0.05);In the positiveexpression of lymph node metastasis group was58.82%lower than the ones withoutlymph node metastasis positive expression rate, which was91.30%, the differencewas significant (P <0.05). Dukes stage A+B of the positive expression rate of88.00%, C+D of positive expression rate was53.33%, difference between the twogroups was significant (P <0.05). Claudin-5is not correlated with gender, age andtumor size.(P>0.05).3. In colorectal carcinoma of Claudin-1and Claudin-5expression and degreeof differentiation of colorectal cancer, expression in dukes staging, lymph nodemetastasis group was negatively correlated, and Claudin-1,Claudin-5have nocorrelation with patients’ gender, age and tumor size.(P>0.05).【Conclusion】1. Claudin-1in normal mucosa, colorectal adenomas, colorectal cancer, hasexpressed, but expression was significantly increased, suggesting that they are maybe associated with colorectal carcinogenesis and progression in colorectal cancer.Colorectal cancer with clinical and pathological features: Claudin-1positive expression and degree of differentiation of colorectal cancer, lymph node metastasisand Dukes stage are correlated.(p<0.05), Claudin-1positive expression has nocorrelation with gender, age,(p>0.05)2. Claudin-5in normal colorectal mucosa, colorectal adenomas and colorectalcancer have expressed. In normal colorectal tissues than in adenomas and colorectalcancer, suggesting that of lower.Claudin-5in colorectal cancer related to incidenceand development of colorectal cancer. Colorectal cancer and the clinical andpathological features: Claudin-5expression and degree of differentiation ofcolorectal cancer, expression and lymph node metastasis and Duke stage arecorrelated (p<0.05), but is not significantly correlated with age, gender and tumorsize.(p>0.05)3. Colorectal cancer in Claudin-1and Claudin-5expression and degree ofdifferentiation of colorectal cancer, lymph node metastasis and dukes staging ofclosely related, The three joint detection for developing colorectal cancergenediagnosis and therapy.
