AIM:To investigate the effect of intraoperative blood salvage (IOBS) on themorphosis of erythrocytes with atomic force microscopy (AFM) observation.METHODS:Blood samples from Ten ASA I~III patients, aged35-65yearsundergoing orthopaedic surgery under general anesthesia, were collected beforeoperation (T1), Unprocessed blood (T2) and processed blood (T3) in cell saver werealso measured. A Fluorescence microscope (×400) was used to examine deformity oferythrocytes in the samples. An AFM with nanometer resolution was used to examineultra-structures of membrane surface of the erythrocytes in the samples.RESULTS:1. The malformation rate of the erythrocytes in T1samples was the lowest (P<0.05).2. There were significant differences in AFM images and height profile of a singleerythrocyte between salvaged blood and venous blood.AFM images at1μmrange showed that the distribution of the particles on the erythrocyte membranesurface in unprocessed blood in cell saver was not regular and was significantlydifferent from the other blood samples.Compared to the other three samples, Ra(profile arithmetic average error)and Rq (root mean square roughness)of in T2samples was the highest (P<0.05).CONCLUSION:There are significant changes in AFM image of erythrocytes and theultra-structures of the membrane surface in salvaged blood....
