In recent years, with the development of economy and improvement of nationalconsumption, the demand of vinous increased year by year and then grape cultivating andwine processing industry developed rapidly. Meanwhile, the by-products-grape skin residuehas been become an urgent problem what has brought the serious environment pressure by thewine-making process system.The extraction of proanthocyanidins was studied by using organic solvent method,tooptimize the extraction conditions for proanthocyanidins from grape skin dregs, solid-liquidratio, extraction temperature, and concentration of ethanol were employed to design responsesurface analysis with yield of proanthocyanidins as response value based on single factor test.The results showed that the optimum extraction conditions were as follow: solid-liquid ratio1:11.15, temperature55.6℃, and ethanol concentration72.8%. Under the optimal process, theextraction rate of proanthocyanidins reached4.45%. The proanthocyanidins thin layerchromatography detection use V(toluene):V(acetone):V(acetic acid)=3:3:1as a developingagent and V(0.6%potassium ferricyanide):V(0.9%ferric chloride)=1:1as the chromogenicagent. The result showed that the chromatography traces of proanthocyanidins by twicepurification as same as the samples (99%purity).Through the response surface analysis, organic solvents and enzymatic assistedextraction of grape seed oil were optimized, and we also obtained the best conditions fororganic extraction of grape seed oil: crush51.2mesh, the temperature53.2℃, extraction time31.35min. The highest rate was185.39mg·g-1. The optimal conditions for enzymatic assistedextraction of1.635%: the amount of enzyme hydrolysis time was55.8min, the hydrolysistemperature was46.℃3. The extraction of grape seed crude oil forecast based on theregression equation408.41mg·g-1, the actual verified extraction of396.91mg·g-1, it wasabout2.1times of the organic solvent extraction, grape seed oil extraction was significantlyincreased after enzymatic hydrolysis.By using the comprehensive extraction condition from response surface methodology,we have got further extraction of resveratrol, proanthocyanidins and grape seed oil from grapeskin residue, and then obtained the following optimal extraction procedure: grape oil→resveratrol→proanthocyanidins.The study on process of active substances in the grape pomace extraction provided atheoretical basis for the grape processing industrial waste recycling. It not only makes goodeconomic returns for the processing industry but also can effectively reduce the environment pressure. It plays an important role at extension of the chain of the grape industry, turningwaste into wealth, improve resource utilization and promotion of local economicdevelopment.
