[Objective]:To investigate the influence of erythropoietin(EPO) on the homing of endothelial progenitor cells and renal reparation in chronic kidney failure(CRF) rats[Methods]:The model of chronic renal failure was established by a two stage5/6nephrectomy procedure on rats. Then animals were randomly divided into three groups:Control group,CRF group,EPO group. After intervention with EPO with a dosage of50IU/kg by hypodermic injection for6weeks,all rats were executed for blood sample and remnant kidney.blood sample were collected for measuring Scr/Bun and Hb/HCT and for isolating blood mononuclear cells from rat heart blood by the method of density gradient centrifugation.remnant kidney was taken for detecting the expression of the protein and mRNA of EPO/EPOR,SDF-1/CXCR4, Angiopoietin-1/Tie2,SCF/C-Kit by the methods of Immunohistochemistry/Western-blot/Real-time PCR,CD31and VEGF were also detecting by Immunohistochemistry method,HE staining was used to evaluated pathological change of the remnant kidney.[Results]:After treatment of rHuEPO,the expression of CD31/VEGF and the density of glomerulus PTC(peritubular capillaries) were increased significantly(P<0.05); the expression of mRNA and protein of EPO/EPOR, SDF-1/CXCR4, Angiopoietin-1/Tie2,SCF/C-Kit in remnant nephridial tissue were raised significantly (P<0.05); the Scr/BUN/Urinary protein of EPO group were reduced significantly, the Hb/HCT of EPO group were increased significantly,and the capillaries endothelial cell of glomerular and PTC were improved,the pathological damage of remnant kidney were ameliorated.[Conclusion]:It might be through activing the homing factors and homing relevant factors of the remnant kidney to improving the ability of EPCs’ mobilization and homing to the impaired kidney to conducting vasculogenesis that EPO can improve the renal function and ameliorate pathological changes of the remnant kidney.
