| Stem cell which has the ability of self-renewing and replicating and cangrow into various tissues and organs is the major discovery in modern medicine.However, various ethical issues appears. In recent years, the discovery and studyof amniotic fluid stem cells provides a new method which is avoiding theethical issues and has more powerful medical potential. Amniotic fluid is easy toobtain and the acquisition process is safe. Obtained while prenatal diagnosis byamniocentesis or while pregnant women deliveries. These two acquisitionmethods will cause no harm to the fetus and cause minimally invasive to themother. Experiments determined that amniotic fluid stem cells have somephysiological characteristics of both embryonic stem cells and adult stem cells.Experiments show that embryonic stem cells can grow into germ cells in vitroand adult stem cells can grow into oocyte-like cells in vitro. This study wasdesigned to study the ability of amniotic fluid stem cells differentiating intooocyte-like cells and discuss the possibility of amniotic fluid stem cells treatingpremature ovarian failure in cell therapy. Objective1. Investigate the ability of second trimester human amniotic fluid stem cellsdifferentiating into oocyte-like cells.2. Transplant human amniotic fluid-derived stem cells through tail vein, andresearch on the treatment of premature ovarian failure.Methods1. Using specific medium induced a second trimester amniotic fluid amnioticfluid stem cell line, make it differente into oocyte-like cells. Finally,markers were identified.2. SD rats were randomly divided into four groups: group A (normal group),group B (model group), group C (once transplantation group), group D(twice transplantation group). Group A was given normal saline. Group B,group C and group D were perfused with Tripterygium Glucosides tablet for14consecutive days. Then group D transplanted AFS twice on the1st dayand the7th day after gastric perfusion and group C transplanted AFS onceon the14th day after gastric perfusion,. Finally, vaginal smears of rats,serum sex hormone levels and ovarian histological changes were observed.Results1. The amniotic fluid stem cells grow rapidly in the medium proving thatamniotic fluid stem cells can be induced and differentiated into oocyte-likecells which express genes ZP2ã€ZP3and protein Vasaã€ZP2in vitro.2. Model group have ovarian failure phenomenon. Ovarian failure of group Cand group D have been restored. twice transplantation group recoveredbetter than once transplantation group.3. New follicles appeared in ovaries of twice transplantation group.4. The differences of E2among the four groups were statistically significant (P<0.01).Conclusions1. Amniotic fluid stem cells have a strong potential for proliferation andcapability of differentiating into oocyte-like cells.2. Amniotic fluid stem cells have a certain therapeutic effect on prematureovarian failure and improve ovarian function from promoting the ovary togenerating follicles and adjust the level of sex hormones. |
