【Objective】:The experiment study is explore to the treatment of rheumatoid arthritiswith the rabbits. using bone marrow stem cell tissue engineering technology, with saltalginate as a scaffold. The paper is about the method of cure rheumatoid arthritis withtissue engineering,through repair cartilage defect. Based experiment’s data and scientificmethod has offered in the research.【Methods】: New Zealand rabbits were devided into normal group,adjuvants modelgroup and salt alginate scaffolds treatment group. Model group and the treatment groupwere established rabbit adjuvants arthritis model, the normal group rabbit had beeninjected the same amount saline. Take the bone marrow in treatment group rabbit.Bonemarrow stem cells were separated and cultured by percoll method in vitro. Continuouslyculture to P3.Induced bone marrow stem cells were differentiated to cartilage cells, andmake salt alginate scaffolds. Mix the cartilage cells with salt alginate scaffold into theknee-joint treatment group of New Zealand rabbit. The normal group and the modelgroup’s joint were injected in the same amount of saline. After a month later,observe therabbit, take the rabbit’s knee for HE dyeing and immunohistochemistry and observe thethree group.【Results】: The primary culture bone marrow stem cells were been found spindle shape,swirling growth.The differentiate cells is most of polygon,collagen type II and the newblue staining is positive and were detected collagen type II mRNA RT-PCR,induction thatthe cells has become cartilage cells. The cartilage cells were mixed with alginatescaffolds.The mechanical strength of scafflold is20.05g/cm2. Tissue engineeringmorphological result show that:compared with the model group, the treatment groupjoint significantly reduce inflammation, cartilage defects are obviously repair.【Conclusion】: With bone marrow cells as seed cells, after inducing to cartilage cells andmixing with cartilage scaffold were injected in the rabbit which can repair adjuvantsarthritis cartilage defect.
