Objective Due to the serious hazards of schistosomiasis japonica, and its effectivetherapy is quite shortage. Therefore, this experiment by observing the role of cyclosporineA (CsA) against Schistosoma japonicum in vivo, provide some evidences and theory basisfor the treatment of Schistosoma japonicum drug development.Methods64gerbils were divided into8groups named as normal group A, model groupB, treat group C, D, E, F, G and H. Gerbil in groups B, C, D, E, F, G and H were infectedwith120+2cercariae of Schistosoma japonicum. Upon infection1week and4week on C,D, E, F, G and H groups were given three different doses (10,20,50mg/kg·d) by CsAtreatment, on5consecutive days. All gerbils were killed after5weeks. Adult worms werecollected and count, the worm reduction rate was calculated. Pathological ation of gerbilliver and intestinal wall: we made routine histopathological sections, and then observed byHE staining and Masson staining. Scanning electron microscopy (SEM) and transmissionelectron microscope (TEM) observed damage of the body’s surface.Result After CsA treatment, the worm reduction rate is found, compared with the modelgroup, the middle dose group and high dose group was significant different (P <0.05), thelow-dose group was not significant different. HE staining and Masson staining resultsshowed that compared with the model group, the middle dose group and high dose group,the area of liver granuloma is reduced, the number of eggs is declined, the degree of liverfibrosis is gradually reduced; and low-dose group had no significant role. Compared withthe normal male and female body, after the CsA treatment they have extensive corticaldamage generally, epidermis wrinkled and sodden, with a large area peel off and generateda large number of emptybubbles. These was be observed by SEM and TEM. Withincreasing dose, the extent of damage is gradually increased, but similar to the damageeffect of drugs on male and female worms.Conclusion CsA can kill both schistosomula and adults in vivo, reducing the number ofeggs, decreasing the area of the hepatic granuloma, declining the degree of liver fibrosis,so as to reducing pathological damage of schistosomiasis japonica.
