Objective:To investigate the radioiodination labeling methods includingselecting the optimum conditions, and performing quality control, all ofwhich are a basis for further realizing the biological behaviors in vitro andpotential strategies for Radioimmunoimaging (RII) and Radioimmunotherapy(RIT) of Pancreatic cancer.Methods:This study describes a fast method for radiolabeling ofAnti-Muc4 with Na125I to construct molecular probe——125I-Mouse-Anti-Muc4, in which both of NBS and Iodogen were used as oxidizing agents.When other conditions are invariable, the reaction temperature, time, ratio ofNa125I/Mouse-Anti-Muc4,Iodogen/Mouse-Anti-Muc4 and NBS/Mouse-Anti-Muc4 are shifted in order to obtain better conditions. The radiolabelingwas analyzed by thin paper chromatography. Then the radiolabeling productswas eluting by the PD-10 columns, collecting protein peak pipe, deterringradioactive and the radiochemical purity. The stability of the radiolabelingcompounds was tested in vitro under different conditions.Results:The radiolabeling ratio of the labeled compound produced bythe Iodogen and NBS methods were about(81.3±0.97)%,(50.7±0.79)%,respectively. The Iodogen is the ideal oxidizing agents. It was the bestcondition for labeling, the ratio of Na125I/Mouse-Anti-Muc4 was 1.5,Iodogen/Mouse-Anti-Muc4 was 10, with 15 min reaction time at roomtemperature (250C). The radiochemical purity was(95.6±1.26)%. After 48 hours stored in NS or 1% fetal bovine serum, the radiochemical purity was tomaintain about 90%.Conclusion:The molecular probe——125I-Mouse-Anti-Muc4 wassuccessfully constructed for the first time; Iodogen as an ideal method forlabeling Mouse- anti-Muc4 with125I, it can get ideal radiolabeling ratio andradiochemical purity. It can affect the radiolabeling results that differentreaction temperature, time and ratio of Na125I / Mouse-Anti-Muc4, Iodogen/Mouse-Anti-Muc4 and NBS/ Mouse-Anti-Muc4. The radiolabeling ratio washigh, and the labeled compounds were stable. But the immune active,biological behavior and potential strategies for Radioimmunoimaging (RII)and Radioimmunotherapy (RIT) of Pancreatic cancer deserves furtherassessment.
