Objective: To clarify the role of Gastrokine 1 in the formation anddevelopment of gastric cancer, we detected the expression of Gastrokine 1 ingastric cancer and paracancerous tissues and analyzed the relationshipbetween the expression level of Gastrokine 1 and clinicopathologicalcharacteristics of gastric cancer patients.Methods: Gastric cancer and paracancerous tissues of 52 gastric cancerpatients were collected just after cutting off during the operations. Theexpression values of Gastrokine 1 in these tissues were detected by real-timefluorescence quantitative polymerase chain reaction (RT-PCR) andimmunohistochemistry. Meanwhile, the informations consisted of gender, age,carcinoma embryonic antigen(CEA) and carbohydrate antigen 19-9 (CA19-9)level in peripheral blood and helicobacter pylori (HP) value preoperation,tumor’s location, depth of invasion, differentiation, lymph node metastasisand stage of the patients were collected. Finally, the experimental datacombined with clinicopathological informations were analysed.Results: The positive rate of Gastrokine 1 protein in gastric cancertissues was obviously lower than paracancerous ones detected withimmunohistochemistry, which were 5.8% (3/52) and 86.5% (45/52)respectively (P<0.01). The expression level of Gastrokine 1 gene in gastriccancer tissues was significantly lower than paracancerous detected withRT-PCR,between which the average△C T valuedifference was 8.22±5.01 (P<0.01). However, there was no significant relationship between theexpression of Gastrokine 1 gene and clinicopathological parameters includinggender, age and CEA, CA19-9 level in peripheral blood preoperation, tumor’slocation, depth of invasion, differentiation, lymph node metastasis, stage ofthe patients (all P>0.05). Moreover, Gastrokine 1 gene expressed remarkablylower in gastric cancer tissues of HP-positive patients than negative ones, andthe average△C T values were 11.14±5.19 and 7.67±4.18respectively(P<0.05).Conclusion: 1. Both Gastrokine 1 gene and protein expressedsignificantly lower in gastric cancer tissues than paracancerous, which suggestthat Gastrokine 1 might play a significant role in a process of the formationand development of gastric cancer as an anti-oncogene. 2. Gastrokine 1 geneexpressed remarkably lower in gastric cancer tissues of HP-positive patientsthan negative ones, which implied that the expression level of Gastrokine 1gene might be down-regulated by HP infection, and then participate in gastriccancerization. 3. The expression level of Gastrokine 1 gene had nothing to dowith clinicopathological parameters including tumor’s location, depth ofinvasion, differentiation, lymph node metastasis, stage, gender, age and CEA,CA19-9 level in peripheral blood preoperation of gastric cancer patients.
