Objective To investigate the expression of Interleukin-6(IL-6) and Interleukin-10(IL-10) in Aspergillus fumigatus keratitis in corneal epithelial cells of rats, and analysis the effect on anti-fungus immunity.Methods72Wistar rats were divided into2sections at random, including Group A1、B1、C1(6rats in each group) and Group A2-. B2、C2(6rats in Group A2and24rats in Group B2C2). Group Aland A2was blank control group. Group Bland B2was the control group. Epithelium of central cornea were also scraped,and then contact lenses were covered in these groups. Group C1and C2was Aspergillus fungal keratrtis group. After epithelium of central cornea were scraped, aspergillus-laden contact lenses were covered in. The corneas were given HE staining and lignt microscope study. The expression of IL-6and IL-10mRNA in corneal epithelial cells of group B (at4h,8h,16h,24h) were assessed by inmmunohistochemistry and semi-quantitative reverse-transcription polymerase chain reaction(RT-PCR). The expression of IL-6and IL-10in corneal epithelial cells of group B (at24h) were detected by inmmunohistochemistry.Results Histopathologic examination:Light mictoscope indicated that Aspergillus fumigatus keratitis was a kind of inflammation, showing the infusion of neutrocytes in large quantity.HE staining revealed the the invasion of fungal hyphae and conidia in the stroma.2. There were almost no expression of IL-6in the blank control group at any observing points. In the experimental group, IL-6mRNA was detected at4h after infection, upregulated with the time-lapse, and up to peak at24h. The difference was statistically significant compared with blank group at any observing points(P<0.01).3. In normal corneas almost no expression of IL-10were detected. The expression of IL-10showed at4h, then upregulated and peaked at24h. The expression of IL-10showed the statistically signficant difference compared with blank group at any observing points(P<0.01) Conclusions1. Fungal culture and10%potassium hydroxide (KOH) smear verified the successful establish of fungal keratitis mode.2. IL-6experessed in corneal epithelial cells in Aspergillus fumigatus keratitis.The increased expression of IL-6in AFK parallelled with the extent of inflammation. IL-6were sensitive factors in fungal keratitis,and the inflammatory status of the cornea can be evaluated by the levels of IL-6.3. IL-10participates in the occurrence and development of AFK, and expression of IL-10still at a high level at the later period. IL-10may be a protective cytokine and play an important role in the process of cornea anti-damage repair through its anti-inflammation and anti-immune function.
