PART1SERUM OF TGF-p, AND VEGF IN PATIENTS WITH NSCLC AND THEIR SIGNIFICANCEObjective To study serum levels of vascular endothelial growth factor (VEGF) and transforming growth factor beta1(TGF-β1) in patients with human non-small cell lung cancer (NSCLC) and their relation with clinicopathological factors, to explore them in the role of angiogenesis with NSCLC and their significance.Methods Enzyme immunoassay (ELISA) double antibody sandwich methods was used to detect serum levels of VEGF and TGF-β1in58patients with NSCLC before surgery, which selected from2009Jan to2010Dec, and29healthy volunteers. Serum levels were compared in the two groups, the relation between their difference and clinical pathological features was explored, and the correlation between them was analyzed further.Results (1) Serum levels of VEGF and TGF-β1in patients were higher than in controls (P<0.05). All the patients, serum levels of VEGF and TGF-β1were positively correlated with low pathological differentiation, advanced clinical stage and lymph node metastasis (P<0.05):lymph node metastasis groups was significantly higher than the without groups, low differentiation groups was higher than in well-differentiated groups, Stage III-IV of the groups was higher than in stage Ⅰ-Ⅱ groups; but there was no statistically significant difference with patients’Sex, age and pathological type (P>0.05).(2) Serum levels of VEGF and TGF-β1was positively correlated in patients of NSCLC (r=0.68).Conclusions The experiment indicates that there are some inner relations between VEGF, TGF-β1and angiogenesis of NSCLC, which may play a role as the tumor markers of the diagnosis, treatment and evaluation of prognosis in NSCLC, with potential clinical application. PART2EXPRESSION OF HIF-1A, FOXP3, VEGF PROTEIN IN TUMOR TISSUES WITH NSCLC AND THEIR SIGNIFICANCEObjective To study the expression of HIF-la, Foxp3, VEGF protein in tumor tissues of NSCLC and their relation with clinicopathological factors, to explore them in the role of angiogenesis with NSCLC and their significance.Methods Immunohistochemical methods was used to detect the expression of HIF-1α, Foxp3, VEGF protein in the tumor tissues of58patients with NSCLC, selected from2009Jan to2010Dec and20cases of normal lung tissue. The expression of HIF-1α, Foxp3, VEGF protein were compared between the two groups, the relation between their difference and clinical pathological features were explored, and the correlation between them were analyzed further.Results (1) The positive expression rates of HIF-1α, Foxp3, VEGF protein in tumor tissues with NSCLC were67.2%(39/58),53.4%(31/58),65.5%(38/58), higher than those in control groups (P<0.05). All the patients, the positive expression of HIF-la, Foxp3, VEGF protein were positively correlated with low pathological differentiation, advanced clinical stage and lymph node metastasis (P<0.05):those in lymph node metastasis groups was significantly higher than the without groups, low differentiation group was higher than in well-differentiated groups, Stage III-IV of the groups was higher than in stage I-II groups; but there was no statistically significant difference with patients’Sex, age and pathological type (P>0.05).(2) The expression of HIF-la, Foxp3, VEGF protein was positively correlated each other in tumor tissue with NSCLC (0<r<1, P<0.01).Conclusions (1) The experiment indicates that HIF-1α, Foxp3, VEGF were highly expressed in tumor tissue of NSCLC, which may act as the tumor markers of the diagnosis, treatment and evaluation of prognosis in NSCLC, with potential clinical application.(2) HIF-1a signaling pathway plays an important role in the mechanism of angiogenesis of NSCLC. VEGF is a downstream regulate gene in HIF-1a pathway. There was positive correlation among the expression of HIF-1α, VEGF, Foxp3in tumor tissue. Therefore, we speculate that in the mechanisms of angiogenesis in NSCLC, VEGF or Foxp3should be a regulated gene in the HIF-1a pathway, that synergistically promoting angiogenesis and tumor invasion in NSCLC.
