Objective:To analyze the effect of amiodarone on funny current(If) in human atiral myocytes of mitral stenosis of rheumatic heart diseas.Methods:Human atrial myocytes were separated acutely in atrial fibrillation and sinus rhythm cells based on preoperative ECG. The atrial fibrillation cells were divided into no treated and amiodarone(10μmol/L) treated groups. Sinus rhythm cells as sinus rhythm group. Amiodarone group was compared to atrial fibrillation group and sinus rhythm group. Then, Observe the impact of atrial fibrillation cell’s If density caused by amiodarone.Results:funny current (If) density of AM Group is significantly lower than the AF group, whereas, there is no significant difference in If density of AM group and SR group. At the test of-70mV, AF group、AM group and SR group average density of I, were (-42.98±2.77) p A/pF.(-28.76±1.34) p A/pF及(-26.93±1.08)p A/pF. After calculated, at the test potential of-40mV-140mV, AF and AM group differences in current density was statistically significant (P<0.01); AF group and SR group differences in current density was statistically significant (P<0.01); At the test potential of-30mV-140mV, AM group and SR group differences in current density was no significant (P>0.05).Conclusion:If density of AM Group is significantly lower than the AF group, If density of AM Group is close to the sinus rhythm cells, suggesting that amiodarone may be the treatment of atrial fibrillation by blocking If role, while prompt If density increase may be one of the mechanisms of atrial fibrillation.
