Introduction:Aim:Detected the expression of stem cell marker ABCG2in Wilms tumor in order to understand whether it can be applied to the detection of the side population(SP) in this tumor.Methods:1, Collected34cases of Wilms tumor operation specimens from2009January to2011May, by immunohistochemical S-P method (streptavidin-perosidase) detected the expression of tumor stem cell marker ABCG2.2, Collected6cases operation fresh tissue specimens were cultured, and preparated into single cell suspension, then stained with fluorescent dye Hoechst33342, in order to observed with the fluorescence microscopy, and detected SP with the Flow Cytometer. Results:1, About76.4%(26/34) of specimens expressed the ABCG2protein in Wilms tumor;2, In3/6cases observed a small amounts of dyeing shallow stain cells with fluorescence microscopy after Hoechst33342dyeing;3, About0.7%of SP cells were detected by FCS, which could be blocked by verapamil. Conclusion:The expression of ABCG2in Wilms’ tumor may be associated with tumor multidrug resistance; A small number of SP cells detected in WT, in which may contain cancer stem cells.
