ObjectiveBetween skin squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) that both originate from epidermis, all belong to the non-melanoma skin malignancies. The squamous cell carcinoma is aggressive and prone to metastasis, but basal cell carcinoma mainly shows destructive local infiltration and hardly to transfer. There is big difference between these two kinds of tumor basic features.At present, many studies have proved VEGF and its receptor’s expression increased greatly in most real tumor tissues, but there is few researchs show the VEGFR-2expression in SCC and the difference and relativity of VEGFR-1and VEGFR-2expression in SCC and BCC tissues. Ration investigating the VEGF, VEGFR-1, and VEGFR-2protein expression in in squamous cell carcinoma, basal cell carcinoma and normal skin tissues, was studied by S-P immunohistochemistry technique.Comparing the difference among them and the correlation of the three protein expression in squamous cell carcinoma to evaluate whether the expression of VEGF, VEGFR-1, and VEGFR-2protein are related to the malignancy of squamous cell carcinoma. We hope that this finding may provide a theoretical basis for new targets of drug treatment and find valuable indicators for the prognosis of disease. MethodsThe tissues of SCC and BCC were obtained from44patients who under went surgery:included26SCC and18BCC, and10cases normal skin tissue samples acted as control group. Of the26cases of SCC,16cases were classified as well differentiated SCC,10cases were classified as moderately and poorly differentiated SCC. S-P immunohistochemistry method was used to detect expressions of VEGF, VEGFR-1and VEGFR-2. SCC and BCC specimens served as the positive control, and substitution of PBS for the primary antibody served as a negative control. The relationships of the results with clinicopathological data were analyzed by integrated optical density (IOD), and analyzed the data with SPSS17.0system.Results1. Distribution of VEGF, VEGFR-1and VEGFR-2was demonstrated within cytoplasm of tumor parenchymal cell; there was a weak expression in keratinocyte above the stratum basale epidermis and part of vascular endothelial cells.2. In SCC, BCC and normal skin tissue, the expression level of VEGF was (129.052±9.933),(116.067±3.864) and (85.254±12.982); The level of VEGFR-1was (130.481±12.020),(117.453±8.157) and (82.193±9.150); The level of VEGFR-2was (136.604±6.672),(121.535±7.499) and (85.430±9.334). There were significant differences in the expression levels of VEGF, VEGFR-1and VEGFR-2among the three groups (F=83.418,78.137, and169.125, P<0.05).3. In well-differentiated SCC and moderately or poorly differentiated SCC, the expression level of VEGF was (124.492±8.969) and (136.346±6.630); the level of VEGFR-1was (127.076±13.616) and (135.930±6.221); The level of VEGFR-2was (133.573±6.563) and (141.453±3.065). VEGF and VEGFR-2protein were negative correlated with the histological grade of SCC (F<0.05). But VEGFR-1positive expression has no obvious relation with the differentiation of SCC (P<0.05).4. In SCC group, the expression levels of VEGF was positively related to those of VEGFR-1(r=0.773, P<0.05) and VEGFR-2(r=0.676, P<0.05), and that of VEGFR-1was positively related to that of VEGFR-2(r=0.718, P<0.05).Conclusion1. The expression of VEGF, VEGFR-1and VEGFR-2protein are related to the biological characteristics of squamous cell carcinoma and basal cell carcinoma. The fiigh expression of them may be one of the reasons caused over proliferation of the non-melanoma skin malignancies.2. VEGF and VEGFR-2protein were negative correlated with the histological grade of squamous cell carcinoma, but VEGFR-1positive expression has no obvious relation with the differentiation of squamous cell carcinoma.3. VEGF was positively correlated with that of VEGFR-1and VEGFR-2in squamous cell carcinoma. They may promote the angiogenesis in tumor jointly.4. VEGF, VEGFR-1and VEGFR-2may play an important role during the development in squamous cell carcinoma and basal cell carcinoma. The combined detection of their expression in non-melanoma skin malignancies may be one of the judgment standards of diagnosis and prognosis of tumor.
