Objective: This essay is intended to reveal the protective effect of adiponectin on human cardiacmyocytes cultured with high glucose, which will offer a new train of thought to the treatment ofdiabetic cardiomyopathy. In the experiment, human cardiac myocytes are cultured. The activityof cell multiplication, the cell apoptosis, and the changes of the expression of Bax/Bcl-2 mRNAare examined under such situation as high glucose and exogenetic adiponectin at different points.Methods:①The cultured human cardiac myocytes are divided into three groups randomly: thenormal group(D-glucose:5.5mmol/l), the high glucose group(D-glucose: 5.5mmol/l), and highglucose plus adiponectin group(D-glucose:5.5mmol/l+ adiponectin :2.5μg/ml).Cultured HCM in24h,48h,72h respectively under the above mentioned conditions.②The OD value was measuredin each group at the different points in order to detect the influence of high glucose and adipone--ctin on the multiplication of HCM.③Collect the cultured HCM at different points, mixed withAnnexin/PI after centrifuging the cell suspension.The apoptosis rate for each group at diffe- -rentpoints was measured by FCM.④Extract RNA of HCM for each group at the different points, a--nd obtain cDNA through reverse transcription. Add in the primer, then measure the expressionof Bax, Bcl-2 via RT-PCR.Results:①Compared with normal group,the sustainable action of high glucose inhabit multipl--ication of HCM.But cell multiplication increase sharply in high glucose plus adiponectin gro-up.The difference has statistical meaning.(P<0.05)②The cell apoptosis rate of HCM increasedsharply in high glucose group compared with normal group. But apoptosis rate decreased obvio-usly with the treatment of adiponectin. The difference has statistical meaning. (P<0.05)③Theexpression of Bax up-regulated while Bcl-2 down-regulated in high-glucose group. The differ-ence between high glucose group and normal group has statistical meaning. (P<0.05)④The ex--pression of Bax increased while Bcl-2 decreased in high glucose plus adiponectin group. More--over, this effect is time-independent. The difference has statistical meaning compared with highglucose group.(P<0.05)Conclusion:The multiplication of HCM and the expression of Bcl-2 decreased, while the apop--tosis rate and the expression of Bax increased under the environment of high-glucose. However,after the treatment of adiponectin, the cell multiplication increased and the cell apoptosis decrea--sed by means of down-regulating Bax and up-regulating Bcl-2 respectively. It plays a positiverole on diabetic cardiomyphy, hence offers a new way to treat diabetic cardiomyophy.
