| Objective:To observe the expression of Angiotensin Ⅱ (AngⅡ)and Transforming growth factor-β1(TGFβ1) in the renal tissue and discuss the protective effect and possible mechanism of Losartan.Methods:54male Sprague-Dawley rats were randomly divided into normal control group (group NC), diabetes mellitus group (group DM), Losartan group (group L). The experimental datas, such as body weight, the blood glucose were monitored weekly. Other datas were monitored at4th week,8th week and12th week separately, such as the blood glucose, the blood insulin, the quantity of urinary protein in24hours, the renal hypertrophy index(renal weight mg/body weight g),the content of Ang II.And observed the morphological changes of the renal tissue under the light microscope, determined the TGFβ1mRNA of renal tissue by Real Time RT-PCR, the content of TGFβ1protein by immunohistochemistry.Results:①The blood glucose, the quantity of urinary protein in24hours, the renal hypertrophy index, the content of Ang Ⅱ, the TGFβ1mRNA and protein of rats in groups DM, L all increased compared with those in group NC, which were all of statistical significance(P<0.05).②The blood insulin of rats in groups DM, L all decreased, which were all of statistical significance(P<0.05).③The expression of renal Ang Ⅱ and TGFβ1diabetes rats were positively correlated with the quantity of urinary protein in24hours (r=0.798,p<0.01)(r=0.808,p<0.01) and the renal weight index (r=0.813,p<0.01)(r=0.816,p<0.01)④The pathological changes of the renal tissue in group L improved significantly over the changes of group DM under the light microscope.Conclusion:The Ang Ⅱ, the TGFβ1of the renal tissue and the quantity of urinary protein in24hours of diabetes rats increased significantly,which related with diabetic nephropathy closely.The Losartan reduces the expression of Ang Ⅱ and TGFβ1in the renal tissue of diabetic rats, improved the renal hypertrophy and reduced the pathological damage. The Losartan may have synergistically protective effect for diabetic nephropathy. |
