Objective:we establish the animal models of facial-nerve denervation at different periods anduse the computer image analyzer and colorimetric analysis technology to observe the changes oforbicularis oculi muscle cell diameter and the enzyme content at different periods in order toprovide a theoretical basis for the best timing of facial-nerve surgical repair.Methods:The 70 of healthy SD rats, weight (200-250) g,divided randomly into seven groups (for1,3 days, 1, 2, 4, 6, 8 weeks after facial-nerve denervation ) each group are 10:experimentalgroups are the orbicularis oculi muscle (right side),normal control groups are orbicularis oculimuscle in the other side (left) which facial-nerve didn’t denervate.The changes of orbicularisoculi muscle cell diameter and the Ca2+-atpases, Na+-K+-atpases content were observed bycomputer image analyzer and colorimetric analysis at different denervation periods.Results:1. The orbicularis oculi muscle cell diameter variation are not apparent for the groups of 1,3 daysafter facial-nerve denervation. The group of 1 week, orbicularis oculi muscle cell diameterreduced about 25.2%, The group of 2 week,it reduced about 35.6%,The group of 4 weeks, itreduced nearly 45.3%, The group of 6 weeks, it reduced almost 60.1%. The group of 8 weeks,theorbicularis ocul muscle cell diameter are the normal amount of 20.5%.The orbicularis oculimuscle cell diameter have a progressive decline with the time extension after facial-nervedenervation.2. The group of 1,3 days after facial-nerve denervation, the gray value (the left side and the rightside) of Ca2+-atpases, Na+-K+-atpases does not change significantly.have no significantdifference (P > 0.05); The group of 1, 2, 4, 6, 8 weeks after facial-nerve denervation, the grayvalue (the left side and the right side) of Ca2+-atpases, Na+-K+-atpases have significantly changeand have a progressive heighten with the time extension after facial-nerve denervation.(the grayvalue of enzyme actual is the reciprocal of its content , the value more smaller,,the content morehigher). The differences were statistically significant(P<0.05).Conclusion:1. The orbicularis oculi muscle cell diameter have a progressive decline with the time extensionafter facial-nerve denervation.2. The content of Ca2+-atpases have a progressive decline with the time extension afterfacial-nerve denervation.3. The content of Na+-K+-atpases have a progressive decline with the time extension afterfacial-nerve denervation.
